Background:In recent years scholars have started to focus on ischemic heart disease, especially myocardial infarction myocardial cell regeneration and recovery of cardiac function, in which bone marrow stem cell transplantation has been recognized as a more promising approach of repair of myocardial injury in heart failure to correct the feasibility. Bone marrow stem cells have multilineage differentiation potential to become a very important seed cells, but multi-differentiation is a double-edged sword, for example the differentiation of malignant tumors.Objective:To investigate by chemical inducing agent 5-azacytidine-induced and simulated myocardial microenvironment of bone marrow mesenchymal stem cells into myocardial-like cell differentiation in vitro and through observation of security of the myocardial-like cells in cell transplantation.Materials:Take one subject of patients with coronary heart disease with operation of coronary artery bypass grafting with marrow,Take another subject of patients with congenital heart disease associated with tetralogy of Fallot with hypertrophic muscle bundle of the right ventricular outflow tract in children with operation of total correction of TOF, and take the removal of the myocardium about 10g when radical surgery to clear the right ventricular outflow tract.12 of BAL-B/C mice of clean grade were randomly divided into cell transplantation group by 5-aza-induced cardiomyocyte-like cell and cell transplantation group by the cell lysate induced cardiac-like cell,6 rats/group.Methods:Take the third generations of bone marrow-derived mesenchymal stem cells treated with chemical inducing agent 5-azacytidine induction and preparation of simulated myocardial cell lysis in vitro induction of myocardial microenvironment 21d; will be a kind of myocardial-like cells. Those were made of 3.0×10/L single-cell suspension and took 0.3ml inoculated by 5-aza-induced myocardial-like cell transplantation after cardiac transplantation group and the cell lysate induced by myocardial-like cell transplantation group, after 3 weeks subcutaneous local tissue specimens from and myocardial tissue contrast observed in nude mice.Results:5-azacytidine and myocardial cell lysate simulated cardiac micro-environment can both induce bone marrow-derived mesenchymal stem cells differentiated into myocardial-like cells in the experimental group than in the control group statistically significant difference (P<0.05), between two experiment groups were no significant difference (P> 0.05) myocardial-like cells in transplanted into the subcutaneous tissue of nude mice in the two groups were not found in malignant cells, immunohistochemical determination of two groups of mesenchymal cell markers Vimentin and cardiac-specific markers CX43 and Troponin T were positive.Conclusion:1 MSCs have the potential to differatiate into myocardial-like cells in suitable conditions.2. MSCs can differatiate into myocardial-like cells with being induced by contact with 5-Aza-induced in vitro and myocardial cell lysate as simulated myocardial microenvironment,and show "environment-dependent divide" characteristics.3 MSCs differentiation into myocardial-like cells transplanted in vitro,and transplant into the nude mice. Through the observation of the short term it does not find tumorigenicity, and has the cardiac migration and homing ability. |
