The ripe dried seed of Oroxylum indicum (L.)Vent. is a kind of Chinese traditional medicine. Most of the relevant literatures abroad and domestic of flavonoids from Oroxylum indicum were studying about the pharmacology, extraction, separation and identification of them, and alcohol-refluxing method was used mostly to extract, while the process optimization of extraction technology and ultrasound-assisted method used to. extract total flavonoids of Oroxylum indicum have not. been reported. Only the content of baicalin is determined on HPLC or RP-HPLC in Oroxylum indicum and Liang-liang Chen and his partners determined the content of index component in Oroxylum indicum with self-made chrysin-7-O-βglucuronopyranoside as a standard, but the content determination of total flavonoids in Oroxylum indicum have not been reported yet. Until now, only 8 fatty acids in seed fatty oil of Oroxylum indicum were separated in a few of foreign literatures. This paper attempts to make a systematic research on the flavonoids and the seed oil of Oroxylum indicum, including establishing the determination method, choosing the extraction method and optimizing the extraction process of total flavornoids, the extraction method and the fatty acid compositions of the seed oil were also investigated in this thesis.1. In this thesis, an UV spectrophotometry method with baicalin as a standard was established to determine the content of total flavonoids from Oroxylum indicum. The standard curve was obtained and its regression equation is C=15.914-A+ 0.1447-x, R2=0.99961. It was informed that this method can be used to determine the content of total flavonoids, which has good linear relationship, high precision, reliable stability and high recovery rate.2. We obtained the extraction process of total flavonoids from Oroxylum indicum. Through comparing the affection of different solvents on extraction rate of total flavonoids, ethanol was chosen as the best solvent. By comparing the effects of Soxhlet method, thermo-refluxing method and ultrasound-assisted method, ultrasound assisted method was chosen as the best one. Single-factor analysis and orthogonal experimental design (L9(34)) were used to test the effects of some operating parameters such as ethanol concentration, solid-to-liquid ratio, extraction temperature, extraction time, ultrasonic power, ultrasonic frequency on the yield of total flavonoids from Oroxylum indicum. The optimal conditions were obtained as follows:ethanol concentration was 70%, ultrasound power was 210 watt, ultrasound frequency was 28 kHz, extraction temperature was 40℃, extraction time was 50 min, solid-to-liquid ratio was 1:30 (g/ml) and extraction times was 1 time. The average extraction rate was 23.829%. It is showed that the extraction process is stability and is able to be carried out.3. We established an extraction method of Oroxylum indicum seed oil. After comparing four extraction solvent, petroleum ether was selected as the best solvent. By comparing the extraction rate and quality of the oil obtained by four extraction methods, Soxhlet method was considered as the best one, and the fatty oil extracted by which was clear and light yellow and its extraction rate was 22.85%, higher than ultrasonic method (20.61%) and microwave method (21.15%).4. Then we established the analysis and identification method for fatty acids in Oroxylum indicum seed oil. After saponified by alkali and methylated by BF3/ methanol, the oil was analyzed by GC-MS and 21 components were identified. It is indicate that the main compositions of the fatty acids in Oroxylum indicum seed oil are oleic acid (57.480%), palmitic acid (9.418%), docosanoic acid (6.882%),11-eicosenoic acid (8.531%), linoleic acid (4.053%), tetracosanoic acid (3.945%), erucic acid (2.934%), stearic acid (1.741%), a-linolenic acid (1.459%) and eicosanoic acid (1.361%), and the total content of unsaturated acids is 76.310%.The result suggests that Oroxylum indicum seed oil has a high developing potential.
