| Oroxyli Semen is the seeds of Oroxylum indicum (L.) Vent. It is largelycultivated in Yunnan, Guangxi, Guizhou Provinces of China. It has the effect ofclearing away the lung-heat, relieving a cough, soothing the liver and regulating thestomach. Pharmacological studies show that, Oroxyli Semen has the effect ofanti-cough, anti-inflammatory, anti-bacterial, anti-cancer anti-oxidant and so on.Due to the reports of the literature, the main chemical components of OroxyliSemen are flavonoids and their glycosides, also containing hydroxyl benzene ethanoland cyclohexanol, volatile oil and so on. During former experiment, two flavonoidshave been extracted and identified as Oroxin A and Oroxin B from Oroxyli Semen,and the contents of these two flavnoids were proved to be very high. In order to makeOroxyli Semen flavonoids development into new drugs, the extraction method,purification process and quality standards of the Oroxyli Semen total flavonoids(OSTF) have been studied in this paper.1. The extraction method of total flavonoids from the seeds of Oroxylum indicum (L.)Vent.This Article use Oroxyli Semen as raw material and two main flavonoids OroxinA and Oroxin B as indexes, the three main influence factors including the extractionsolvent, amount of extraction solvent and extraction duration were designed inorthogonal tests. Through the experiment, the optimal extraction process wasdetermined as using8times of60%ethanol (V/W) refluxing extraction for4times,extraction time were60,45,30,30min, respectively.2. The purification process of total flavonoids from the seeds of Oroxylum indicum(L.) Vent.This paper analyzes7different types of macroporous adsorption resin (D4020,AB-8, HPD100, DM301, DM130, D1400, D101), the static adsorption, desorptionand other properties were studied, and the results show that HPD100macroporousadsorption resins for Oroxyli Semen is better than others. So this paper choice theHPD100macroporous adsorption resins for SOTF. Liquid concentration, solution pHvalue, liquid flow rate as the three main influence factors were investigated, todetermine the HPD100macroporous resin adsorption conditions. And then the HPD100macroporous adsorption resin desorption conditions were studied,determined the eluent concentration and dosage.According to the above results, ultimately determine the purification methods ofOSTF are as follows.Extract the Oroxyli Semen powder as the optimal extraction process, mergingextracted liquid, filtering after decompression and concentration to no alcohol taste,diluted with water to the volume of the medicine20times. Regulate the pH to5.0,after the adsorption by HPD100macroporous resin at3BV/h, distilled water wash tocolorless,60%ethanol elution, eluent decompression and concentration to no alcoholtaste, dried, then obtained the OSTF. By HPLC, the content of Oroxin A and Oroxin Bachieves60%above, the transfer rate of Oroxin A and Oroxin B were more than80%.3. Studies on chemical constituents of Oroxyli Semen total flavonoidsIn this paper, the OSTF in chemical composition was isolated by preparativeHPLC and identified by HPLC,13C NMR,1H NMR and other means. Several majorchemical constituents were identified as Oroxin A, OroxinB, Chrysin-7-O-diglucoside,Baicalin,Chrysin-7-O-β-galactopyranuronoside and Baicalein.4. The quality standard of total flavonoids from the seeds of Oroxylum indicum (L.)Vent.This paper also examines part of the quality standards of OSTF, completed theidentification, content determination, the loss on drying, residue on ignition and seriesof research work. In addition to establish content determination method of the2mainflavonoids Oroxin A and Oroxin B in OSTF by HPLC, multiple batches of sampleswere determined. The experimental results show that Oroxin A content the mean of27.14%and Oroxin B content averages38.19%in OSTF. The contents of these twokinds of flavone glycosides, Oroxin A and Oroxin B in SOTF were studied and theresults showed that the method of determination of the content were determination ofwide linear range, accuracy, precision, sensitivity is very good, so it can be used as thetwo flavonoids contents determination method.5. The DPPH radical scavenging activityThe experimental results showed that, the OSTF, Oroxin A and Oroxin B havethe ability of scavenging capacity while Chrysin-7-O-diglucoside can’t. The Sequenceof the DPPH radical scavenging activity as: Oroxyli Semen total flavonoids>OroxinB> OroxinA> Chrysin-7-O-diglucoside. In conclusion, this paper undertook the system studies on Oroxyli Semen totalflavonoids preparation process, quality standards, chemical composition and theDPPH radical scavenging activity. The preparation process is convenient, low cost,high yield, environmental protection and suitable for industrialized production. Theresults provide pharmacy and pharmacodynamic basis for Oroxyli Semen totalflavonoids to be developed into a new drug. |
