| Objective:To strengthen the research of the rat model of intrauterine device bleeding with Qi-deficiency-blood-stasis syndrome, to examine the characteristics, etiology and pathogenesis of intrauterine device bleeding with Qi-deficiency-blood-stasis syndrome from a deeper perspective and level, and to provide some new treatment methods and ideas of this disease.Methods:24 SD rats were divided into 4 groups:blank control group, implant ring group, model group and treatment group,each group has 6 rats.Except blank control group, the other 3 groups rats were implanted ring, model group and treatment group were built the the rat model of intrauterine device bleeding with Qi-deficiency-blood-stasis syndrome by compound methods for 28 days.From the 1st day of molding, the rats were on diet, forced to swim. And from the 14th day, they were hypodermic injected adrenal hormones.The treatment group was treated with intrauterine device No.Ⅱprescription from the 21st day for 7 days. The general behaviors,body weight changes, uterine morphology smears and various indicators of protein chips of the 4 groups rats were observed and analyzed. Results:The model group rats appeared Qi-deficiency, fatigue, loose stools and other performances, and their body weight significantly reduced compared with pre-made model (P<0.01),the uterine tissue morphology smears showed epithelial cells, necrosis and inflammatory cell infiltration. Among the indicators in protein chip of the uterine tissues, the relative expression level of IFN-y and IL-4 in model group reduced obviously comparing with that of blank control group and implant ring group(P<0.01), which in treatment group increased comparing with that of model group (0.01<P<0.05).The relative expression level of MIP-3a in model group reduced obviously comparing with that of blank control group(P<0.01),while compared with that of implant ring group had no statistically significant(P>0.05),which in treatment group increased comparing with that of model group(0.01 0.05)comparing with that of blank control group and implant ring group, which in treatment group reduced comparing with that of model group(0.01 0.05)Conclusion:After being implanted ring, on diet, forced to swim and hypodermic injected adrenal hormones,the rats'general behavior and body weight have been changed. And the uterine histology, indicators in protein chip have also changed. These all prove that we have successful built the rat model of intrauterine device bleeding with Qi-deficiency-blood-stasis syndrome. |
