| Objective:To observe the effect of Angelica polysaccharide extracted from Gansu genuine Chinese angelica root on changes of pulmonary function, cellular morphology, extracellular matrix and etc. in lung fiberized rats. To provide the clinical foundation for both prevention and treatment of lung fiberosis.Methods:60 healthy Wistar rats were randomly divided into six groups: blank control group, positive drug (dexamethasonum) control group, high, medium, low dosage of Angelica polysaccharide group with 10 rats in each group. Except blank control group, other groups were replicated to rat pulmonary fiberosis model induced by bleomycin at dosage of 5mg/kg by using trachea injection. On the second day after modeling, high, medium, low dosage groups were intragastricly administered with Angelica polysaccharide at 200mg/kg, 100mg/kg,50mg/kg per day respectively. The positive drug group was intragastricly administered with suspension of dexamethasonum acetate at lmg/kg daily. The blank control group and model group were intragastricly administered with isometric saline at 10ml/kg. All groups were administered for 28 days continuously. After 1 hour of administration on the 28th day, the rat of each group was anaesthetized with 3% napental 40mg/kg by abdominal injection. After 5-8mins, it was regularly positioned and inserted with trachea catheter by thrachetomy and then the rat was dorsal ly placed in volume scanning chamber of animal respirator. When it breathed, the dilatation and contraction of rat's pleural cavity would change volume pressure of chamber in which the change of lung volume could be obtained collaterally from variation of volume pressure. The index of pulmonary function could be obtained by computer. As rat was weighed and it's lung tissue obtained, the lung index were calculated by comparison of weight between body and lung. Following results were made by certain methods:the pathological change observed under microscope, content of hyaluronic acid(HA) and laminin(LN) examined by enzymoimmunoassay as well as hydroxyproline (Hyp) by spectrometer.Results:Compared with normal control group, experimental results were the apparent increase of lung index, airway resistance and the decrease of lung compliance, FVC, percentage of FEV0.3/FVC, FRC and Lung index in model group, but there were great change of pathological morphology character in lung fiberized rats under micrope and content of HA, LN and Hyp of lung tissue increased in model group to some degree. Compared with model group, in groups of Angelica polysaccharide and dexamethasonum experimental results were the obvious reduction of lung index, degree of pulmonary fiberosis, airway resistance, but the increase in lung compliance, FVC, percentage of FEV0.3/FVC, FRC to some degree, angelica polysaccharide has made great improvement in the change of pathological morphology character and content of HA, LN and Hyp of lung tissue were decreased some degree.Conclusion:Angelica polysaccharide has made great improvement in the changes of pulmonary function, pathological morphology, extracellular stroma metabolism and etc. in lung fiberized rats. |
