Objective:To observe the influence of Fufang Danshenyin on the nitric oxide(NO), endothelin-1(ET-1) and body weight, blood pressure,glucose and lipid metabolism of the vascular endothelial cell (VEC)in rats with insulin- resistant (IR),and to explore the protective mechanism of vascular endothelial function.Methods:60 clean male wistar rats were randomly divided into 5 groups, 12 in each group:control group, model group, positive control group, traditional Chinese medicine therapy group,traditional Chinese medicine prevention group.We adopted the model which High fat high carbohydrate diet induced insulin resistance and insulin resistance. We continued feeding all rats for 8 weeks, after the IR model being establishment, then all rats were killed. We adopted the method of biochemical enzymatic to test triglyceride (TC),cholesterol,high density lipoprotein (HDL),low density lipoprotein (LDL), and the method of enzymelinked immunosorbent assay to measure NO, ET-1 levels.Results:1 After 8 weeks:compared with the control group,other groups with high-fat high-carbohydrate diet increased levels of fasting serum insulin (P<0.05),There was statistical significance;sensitivity index decreased (P< 0.01),there was statistical significance;body weight,blood glucose, blood pressure levels were elevated in other groups (P<0.05),with statistical significance.It stated that we achieved to copy the model of insulin resistance.2 After 16 weeks:â‘ Compared with the control group:Chinese medicine prevention group reduces NO, increased ET-1(P>0.05), there was no statistical significance between them;the other three groups decreased NO, increased ET-1(P<0.01),with statistical significance.â‘¡Compared with model group:the contents of NO increased and ET-1 reduced in positive control group,traditional Chinese medicine therapy group and prevention group (P<0.05),with statistical significance;and weight,blood pressure,blood sugar, blood lipids were changed (P<0.01),with statistical significance.â‘¢Compared the positive control group:the contents of NO increased and ET-1 reduced in Chinese medicine therapy group (P>0.05),there was no statistical significance between them;and weight increased,blood pressure decreased in Chinese medicine therapy group (P<0.05),with statistical significance; blood glucose, blood lipid changes (P>0.05),without statistical significance. the contents of NO increased and ET-1 reduced in the prevention group (P<0.05),with statistical significance;other indexes had changed (P>0.05),without statistical significance.Conclusion:1 Fufang Danshenyin can increase the contents NO and reduce the contents of ET-1 in rats with IR. It shows that Fufang Danshenyin has the protection of vascular endothelial function.2 Fufang Danshenyin can prevent insulin resistance,to suggest that it has the effect of prevent the the injury of vascular endothelial cell injury.3 Fufang Danshenyin has an effect of adjusting lipid, reducing blood pressure,improving obesity and disturbance of glucose metabolism.
