ObjectiveTo investigate the tyrosine phosphorylation and protein expression of insulin receptor substrate-1 in Endometrial tissue from patients with preeclam-psia (PE).To study the pathogenesis To provide support in theory for prevention of PE and reduced the mortality of PE patients and newborn.MethodsChose 30 preeclampsia inpatients 15 of severe preeclampsia and 15 of mild preeclampsia)of late pregnancy of Jinan municipal women and children's hospital,and 30 normal late pregnancy women as the control. Study conditions: Select without liver, kidney disease, non-essential hypertension, hereditary diseases. Fasting insulin (FIN) Was measured by radioimmunoassay. Fasting plasma glucose(FPG)Was measured by oxidase assay. Insulin resistance index was calculated using homeostasis model assessment(HOMA)to analyze the relationship between these markers and insulin resistance.The expression of IRS-1 and phosphorylation of Tyrosine phosphokinase were analyzed by immunohistochemistry.Results(1) The age, height, weight of the preeclampsia group and the normal prognancy group were not statistically. (2) The levels of FINS and insulin resistance index were calculated according to homeostasis model assessment in preeclampsia group were significantly higher than those in normal prognancy group (P<0.01). The levels of FPG was no statistical meaning(P>0.05)。(3) The level of IRS-1 protein expression in preeclampsia Group was lower than that in normal pregancy group (P<0.01)。(4) The tyrosine phosphorylation in preeclampsia Group was lower than that in normal pregancy group (P< 0.01).(5) Protein expression and TP with IRS-1 was negatively related to HOMA-IR in preeclampsia group(r=-0.550584,P<0.01)。Conclusion(1) Changes in protein expression and tyrosine phosphory lation of IRS-1 in endometrial may be one of the molecular mechanisms leading to insulin resistance in patiant with preeclampsia.(2)The levels of FINS and HOMA-IR in preeclampsia group were significantly higher than those in nomar pregnancy group. Tips for women with pre-eclampsia there is insulin resistance. (3) The author suggested that the further research of Increased IRS-1 protein expression and tyrosine phosphorylation of insulin resistance to lower levels be helpful for the prevention and treatment of the hypertensive disorder complicating pregnancy.
