Objective:1. The heat research of prevention and treatment of PVR was to use durg to inhibit RPE cells proliferation at present.In this study,inhibition effect of Tet-5-FU-CHT-MS on proliferation of rabbit retinal pigment epithelial cells in vitro and its mechanism were studied.2. the effectiveness of curcumin on the prevention and treatment of experimental proliferative vitreoretinopathy(PVR)in rabbit eyes was evaluated.And the Anti-inflammatory mechanisms of Tetrandrine was studyed.Methods:1. The RPE cells were isolated and collected from the pigmented rabbit and cultured and passaged in 10%FBS-DMEM and identified by anti-human keratin.Cultured RPE cells were divided into 2,5,10,15μg/mL Tet group and 5,10,15,20μg/mL 5-FU group,the blank control group according to the utilization of drug.The MTT assay was used to evaluate the inhibition effect of different doses of indifferent time.Cell cycle,mitochondrial transmembrane potential(Δψm),Ca2+ and apoptosis of RPE cells cultured with 10μg/mL Tet and 15μg/mL Tet were detected using flow cytometry in defferent time.2. The PVR model was induced by 48 mature New Zealand albino rabbits(96eyes) randomly divided into three groups.One group has 16 rabits(32 eyes).0.2ml BSS of Tet-5-FU-CHT-MS was injected in A group,0.2ml BSS Of 5-FU-CHT-MS was injected in B group.0.2ml was injected in C group,this group was taken as control group.Changes of cornea were examined and recorded by slit lamp biomicroscope indirect ophthalmoscope,funduscolor camera and B ultrasonograph after injection each day.The effectiveness of Tet-5-FU-CHT-MS was evaluated by the incidence rate of retinal detachment. On 7,14 and 28d after injection Vitreous humor of each group was taken 0.2ml, detected by enzyme-linked immunosorbent assay (ELISA) in TNF-a. IL-1 content.Results:1 RPE cells were significantly inhibited by Tet and 5-Fu in a dose-dependent and time-dependent manner, there was significant difference comparied with control group(P<0.05) except Tet 10,15μg/ml and 5-Fu 15μg/ml,20μg/mK 15μg/mL at 96h. Cell cycel analysis indicated that Tet and 5-FU blocked cultured RPE cells in G0/G1 phase.The apoptosis rate of RPE cells was significantly higher at 8,24,48 and 72 hours after cultured with 10(μg/mL of Tet and 15μg/mL 5-Fu, comparied with control group(P<0.05),Δψm was significantly decreased than that of the control group respectively(P<0.05). Ca2+ was significantly increasedthan that of control group respectively(P<0.05).2. The rate of the retinal detachment was significantly decreased than the Other groups(P<0.05). TNF-a, IL-1 content of each vitreous humor was significantly decreased than the Other groups(P<0.05).Conclusion:1.Tet and 5-FU obviousiy inhibits the proliferation of rabbit RPE, induced the RPE apoptosis.2.Tet-5-FU-CHT-MS has a good anti-inflammatory effect in eyes, in prevention of proliferative yvitreoretinopath.Tet-5-FU-CIIT-MS can effectively inhibit the occurrence and development of proliferative yvitreoretinopath in rabbit model.
