Objective:Our aim is to detect the relationship between protein location & expression levels of ERaand ERβin correlated region of hippocampus & hypothalamus of Premenstrual Syndrome model rats with liver-qi depression, which gives a preliminary discussion on the pathogenesis of PMS with liver-qi depression,as well as studies the effects of Jingqianshu granule.Methods:After being chosen by open field test,macro-behavior observation and vaginal smear examination,PMS model rats with liver-qi depression were prepared by using bondage stimulation.And then the rats were evaluated by open field test and macro-behavior observation.We detected the protein location,integrated optical density(IOD) in CA1 & CA3 of hippocampus and in ventromedial hypothalamic area(VMH)of rats in the groups including the normal group,bound model group,and treatment group by using immunohistochemistry ABC methods,and calculated IOD(ERβ)/IOD(ERa).Results:Compared with the normal group,the weight of rats in bound model group lessened significantly(P<0.01),and the crossing score,rearing score and total score of open field test all decreased obviously(P<0.01).Meanwhile, the rats showed listlessness,dull in eyes and insensitive to surrounding stimulation.On the other hand,compared with the bound model group,the weight of rats in treatment group increased significantly(P<0.05),and the open field scores all increased remarkably(P<0.05 or P<0.01).IHC demonstrated that there was no noticeable differences in location of ERaand ERPin hippocampus and hypothalamus of rats in three groups.Compared with the normal group,the shape of pyramidal cell in CA1 & CA3 of hippocampus changed,and the numerical value of IOD of ERaand ERPincreased markedly (P<0.05 or P<0.01)in CA1 & CA3 of hippocampus and in VMH,and there was no statistical differences in IOD(ERβ)/IOD(ERa) in the model group. There was no abnormal change in hippocampal formation of rats in treatment group.The numerical value of IOD of ERaand ERβdecreased evidently (P<0.01)in CA1 & CA3 of hippocampus and in VMH,and IOD(ERβ)/IOD(ERa) decreased remarkably(P<0.05)in VMH compared with the model group.Conclusion:The model rats with PMS liver-qi depression can be prepared successfully by using bondage stimulation,and they can be evaluated effectively by open-field test and macro-behavior observation.The high expressions of the ERaand ERβprotein in CA1 & CA3 of hippocampus and in VMH may have close relationship with PMS liver-qi depression,and probably is one of the centre pathogenesis of PMS with liver-qi depression.The Jingqianshu granule can correct the high expression of ERaand ERPprotein of model rats in above brain regions,as well as down-regulate ERβ/ERa in VMH,which may be one of the mechanism to treat PMS with liver-qi depression.
