Objectives:Our aim were to seek for partial micro-mechanism of premenstrual syndrome liver-qi depression by measuring the special ligand binding of estrogen receptor P(ERβ)in different brain regions and detecting the estradiol level in the serum, and study the effect on Jingqianshu granule acting on the liver-qi depression.Methods:PMS liver-qi depression has been introduced by bondaging the limbs in rats,and some of rats were treated with Jingqianshu granule.Then,we observed cell shape by using the method of vaginal smear,and judged the estrum of rats:receptive phases and non-receptive phases.Subsequently, we evaluated the animal behavior by open-field test.After that,we killed the rats by decapitation,got mixed blood and dissected hippocampus,hypothalamus,frontal cortex and parietal cortex from the brains and then prepared the cytosol.The ligand binding activity of estrogen receptor P in different brain areas were detected by radioligand-receptor binding techniques,The content of estradiol in the serum was examined by the radio-immune assay.Results:The macroscopic ethology results showed that:the body weight of the model group rats decreased significantly(P<0.05) compared with the normal group.Compared with the model group,the body weight of rats increased obviously(P<0.05)after treating with the Jingqianshu graulne.Compared with the normal group,the crossing score, the rearing score and the total score of the bondage group rats in open-field decreased markedly(P<0.05).After treatment with the Jingqianshu graulne, those there scores in open-field increased statistically (P<0.05). Compared with the normal group, the specific binding activity of ERβin the hippocampus, hypothalam-us,frontal cortex and parietal cortex all increased significantly (P<0.05)in radioliga-nd-receptor binding experiment.Compared the model group,the specific activity of the hypothalamus,frontal cortex and parietal cortex all decreased signifycantly(P<0.05). Compared with the normal group, the content of estradiol in the serum decrea-sed remarkably (P<0.05). Compared the model group, the estradiol levels increased evidently after treating with the Jingqianshu graulne.Conclusions:Our results sugest there is a intimate relationship between the bin-ding activity of ERβin different brain regions,the serum estradiol level and the PMS liver-qi depression.The binding activity of ERP in the hippocampus,hypothalamus, frontal cortex and parietal cortex significantly up-regulated and the decreased estradiol levels in the serum may be important reasons that cause PMS liver-qi depression,and Jingqianshu granule may cure the diseases by regulating the above abnormal changes.
