The Effects Of L008 On Serum C-Reactive Protein And Aortic Morphology Of Rats At The Early Stage Of Atherosclerosis

Objective To study the effect of herbs L008 on serum C-Reactive Protein and aortic morphology in early atherosclerosis rats fed by high fat diet ,and approach that it has the effect of anti-inflammatory and protection on vascular endothelium cell on the experiment atherosclerosis rats.Methods thirty male mature Sprague-Dawley rats were randomized into normal group fed with common food(n=10) and the group with high fat diet. The other group(n=20)-each was fed 25g fatty food from 18:00 to 8:00 the next day - was fed with fatty food for 8 weeks . At the end of the fourth week , the group fed with fatty food was randomized into model group and drug intervention group that was administered L008 (4g/kg.d )via gavage four weeks . Normal saline was given to rats of normal group and model group. Weighing at a fixed time each week, then adjusting the dose according to the amount of body weight . After 8 weeks, the blood was taken out and all rats were killed. TC,TG,LDL-C and CRP were measured at the end of the fourth and the eighth week. Morphological changes of aorta were observed under gross and light microscopes.Results 1. In general, normal group have coat luster, activities freely and lively. The weight of model group fed high fat diet increased rapidly, and subcutaneous fat is thicker, but with the experiment carried out, the group reduce activities and food, and be in poor mental state. In all respects,The drug intervention group was better than the model group. 2. At the end of fourth week, the lipid level of the group with fatty food were higher than the normal group(P<0.05). 3 . At the end of fourth week, the level of CRP in normal group were lower than the other(P<0.05). At the end of eighth week, the levers of CRP could be better controlled in the group administrated drugs(P<0.05). 4 .The changes of the early stage of atherosclerosis in model group were focal or transient areas of endothelial loss, monocyte adhered to endothelium or moved into subendothelia space , proliferation of smooth muscle cell and so on. The pathological changes in the drug intervention group were improved compared with model group.Conclusion 1. The L008 can downregulate the serum levels of CRP. 2. Morphological changes of aorta show a significant effect of protecting aorta endothelial cell,inhibiting monocyte adhering to endothelium and decreasing the proliferation of vascular smooth muscle cell in rats fed with The L008. As a result , The L008 can prevent the progression of atherosclerosis by anti-inflammatory.
