The development, quality control and safety evaluation of less harmful cigarette has been one of urgent problems to be solved in international tobacco industry. At present, the researches and production of the Low-Harmful Cigarette Added with Chinese Medicinal Herb are booming in the industry. Therefore, it should be accelerating the research of its quality control, making a quality standard of the less harmful cigarettes added Chinese herbal as soon as possible and establishing the corresponding safety evaluation system. The object of this study is'jiangshan' is the low-harmful cigarette that contains Chinese herbal extraction, using the FLD-HPLC, DAD-HPLC, GC technology and cell experiment comprehensively to evaluate the quality control and safety of this kind of cigarette, in order to lay a foundation to develop the less harmful cigarettes with Chinese herbal medicine in the future. The main contents and the results of the study as follows:1. It established a GC-FID method to analyze the active components of Salvia, Scutellaria in herbal additives, tobacco leaf, mainsteam smoke and filter tip. Methodological study showed that tanshinone II A and baicalin in the detection limits were 1.2,0.77μg/mL, respectively, tanshinone II A in 2.0-60.0μg/mL, baicalin in 1.55-200.0μg/mL range with a good linear relationship; the extraction recoveries were at 78.6%-97.1%and the recovery rate was between 76.7%-95.1%; within-day precision and reproducibility of the test results well, two index components can reach the baseline separation with the interference component in tobacco leaf, main smoke and filter tip. These results showed that the method was suitable for the determination of baicalin and tanshinoneⅡA study in sample ciaggrettes. Meanwhile, it determined two main components in Salvia, Scutellaria while their trapping transferring rate and the capture of filter transfer rate. The results convey that tanshinoneⅡA content was 6.6μg in one piece, trapping transferring rate was 27.1%and filter retention rate was 25.6%, while, baicalin was 8.5μg in one piece, trapping transferring rate was 20.5%and filter retention rate was 16.5%.2. It established a method to analyze benzo[a]pyrene that was the harmful substances in main smoke, meanwhile studied the effect of Chinese medicine extraction on the content of benzo[a]pyrene in mainstream smoke. Methodological study showed that benzo[a]pyrene detection limit was 9.1 ng/mL; in 12.5-410.0 ng/mL range has a good linear relationship; the high, medium and low extraction recoveries was between 90.0%-96.4%, reproducibility and stability results well; benzo[a]pyrene and interference components in mainstream smoke can reach the baseline analysis. These results showed that the method was suitable for benzo[a]pyrene determination in mainsream smoke in sample cigarettes. Application of the method to study the effect of Chinese medicine extraction on content of benzo[a]pyrene in mainstream smoke, the results showed that compared with the control cigarettes, cigarettes adding Salvia or Scutellaria extraction had less harmful components of benzo[a]pyrene in mainstream smoke, decreased 30.2%, 13.4%in mainsteam smoke,25.2%,15.5%in filter tip respectively.3. To study the effect of tanshinoneⅡA and baicalin protect vascular endothelial cells that injuried by benzo[a]pyrene. MTT method results showed that 50μmol/L of benzo[a]pyrene can reduce the formation of Formazan in CRL-1730 cells and decrease the OD values. Compared with the control group was a significant difference(P <0.01).While pre-added baicalin and tanshinoneⅡA can promote the formation of Formazan and inhibit benzo[a]pyrene to injury CRL-1730 cell. When the dose of tanshinoneⅡA and baicalin up to 15μg/mL,30μg/mL respectively, they had a significant protective effect (P<0.05). The result that tanshinoneⅡA and baicalin effects on the MDA, LDH, SOD level found that 50μmol/L of benzo[a]pyrene can increase MDA and LDH released, and decreased SOD activity. While pre-added tanshinoneⅡA and baicalin into the cell can decrease MDA and LDH released, and increased SOD activity. TanshinoneⅡA and baicalin have significant protective effect on CRL-1730 cell when reach 15μg/mL,30μg/mL respectively.
