Objective To examine the expression of hypoxia-inducible factor 1 alpha (HIF-1α), glucose transporter protein -1 (Glut-1), Ki-67 at different stages of hemangioma and vascular malformations and its significance.Methods Collected 45 cases with hemangioma by surgical resection in department of pathology, the first affiliated hospital of Guangxi Medical University, between January 2005 and December 2009, grouped by pathological section of hematoxylin-eosin staining (HE) staining, 10 cases of kids'skin and soft tissues as normal control group. 4μm thickness of all specimens were serially sectioned, routine dewaxing and hydration, treatmented by immunohistochemical (IHC) streptavidin biotin - peroxidase method link (SP), fixed antigen through high temperature and pressure, each section in the high magnification (400×) were randomly selected sample of 5 horizons, observed the expression of HIF-1α, Glut-1,and Ki-67 by counting the number of positive cells.Results Specimens of 45 cases of hemangioma by Mulliken classification standards were divided into 30 cases of hemangioma, including 19 cases of proliferating stage(aged 3 months to 1 year old), 11 cases of lnvoluting stage(aged 10 months to 9 years of age), 15 cases of vascular malformations(age 7 years to 52 years). HIF-1αpositive staining cells was localized in the cytoplasm or nucleus, brownish yellow; proliferative hemangiomas in the cytoplasm and the nucleus of a large number of brown-yellow granules, and involuting hemangiomas, vascular malformations and normal control group, only a small brown particles; statistical results showed that hemangiomas in the proliferative phase was significantly higher than that of HIF-1αinvoluting hemangiomas, vascular malformations and normal control group, the difference was statistically significant (P<0.05). Glut-1 positive staining cells localized in the membrane of vascular endothelial cells, or cytoplasm, brownish yellow; proliferative hemangiomas cell membrane in a large number of brown-yellow granules, involuting hemangioma endothelial cell membrane a small amount of brown yellow granules, and vascular malformations and normal control group, endothelial cell membrane only very little or no deposition of brown granules; statistical results showed that hemangiomas in the proliferative phase was significantly higher than that of Glut-1 tumor regression of blood vessels, the difference was statistically significant (P<0.05), Glut-1 were all negative in vascular malformations and normal control group. Ki-67 positive staining cells was localized in the nuclei of endothelial cells, showing brown yellow or brown; proliferative hemangiomas in the nuclei of a large number of brown-yellow or brown granules, and involuting hemangiomas, vascular malformations and normal endothelial cells in the control group, only a small amount of cytoplasm and nuclei of brown particles, statistical results showed that hemangiomas in the proliferative Ki-67 was significantly higher than that of involuting hemangiomas, vascular malformations and normal control group, the difference was statistically significant (P<0.05). HIF-1αand Glut-1 expression was positively correlated in hemangioma (P<0.05). HIF-1αand Ki-67, Glut-1 and Ki-67 expression was positively correlated in hemangioma (P<0.05). Conclusion The abnormal expression of HIF-1αand Glut-1 in hemangioma occurrence and development of a certain relationship, HIF-1αby upregulating the expression of Glut-1 to promote proliferation of endothelial cells.
