| Objective: To investigate the effects of leptin on P450 aromatase of human luteinized granulose cells of PCOS in vitro.Methods: Human luteinized granulose cells from in vitro fertilization-embryo transfer (IVF-ET) are divided into two groups: PCOS group and normal group. They were cultured with medium for 24h after purification, and then cultured with medium plus various concentration of letpin (0ng/ml, 1ng/ml, 10ng/ml, 20ng/ml, 100ng/ml, 1000ng/ml) and FSH (5mIU/ml) in combined with Androstenedione (1μmol/ml) for 24h. The media was collected to measure the concentration of E2 production by immunoradio assay, to measure the concentration of A by enzyme linked immunosorbent assay (ELISA). And then evaluate the activity of P450 aromatase of ovarian granulose cell by the value of E2/A.Results: The estradiol level is decrease with the concentration of leptin increase, and the level of androstenedione is increase with the increasing of leptin concentration, in the culture supernatant in both the PCOS group and the normal group. The difference between the two groups has statistical significance (P<0.05). Whereas the value of E2/A is decrease with the increasing of leptin concentration in both the PCOS group and the normal group, but the difference has no statistical significance (P>0.05). The value of E2/A of normal group is higher than of the PCOS group when the concentration of leptin is 0ng/ml, and the difference has statistical significance (P<0.05).Conclusion: 1. Leptin caused a concentration-related inhibition of estradiol production in luteinized granulose cells of PCOS. 2. The activity of P450 aromatase of luteinized ovarian granulose cell of PCOS is weaker than that of the normal female. 3. High leptin concentration can inhibit the activity of P450 aromatase of luteinized granulose cells, inhibit the aromatization of androstenedione, and prevent the transformation of androstenedione into estradiol, thus causes the increasing of serum androgen level. |
