Objective:1. To optimize the the ethanol extracting technology condition of persimmon leaves flavonoids, and prepare the persimmon leaves flavonoid tablet.2. To study the quality of persimmon leaves flavonoids tablet and establish the method for its quality control.Methods:The optimum procedures in the ethanol extracting technology condition was sleeted by the orthogonal text design L9 (34) respectively. The yield of the quercetin extraction was used as the assessment in it. The content of quercetin was determined by HPLC. The starches, povidone iodine, and magnesium stearate were added after the extractive of persimmon leaves flavonoids were shattered into powder, then persimmon leaves flavonoid tablets were prepared after the mixture was dried and sifted. Persimmon leaves flavonoids were identified by TLC. The contents of quercetin were determined by HPLC.Results:The optimum condition was that the crude drugs were the circumfluence extracting for 3 times with 10-fold ethanol (with 12-fold ethanol for the first time),2 h each time (1.5 h for the last time). The concentration of alcohol was 70%. Persimmon leaves flavonoids were extracted in turn by petroleum ether, and then by ethyl after the tests for recovery alcohol from filter liquor were conducted by using vacuum distillation. The physic liquor was centrifugated, concentrated and dried by microwave vacuum concentration dryer to dry powder. The starches, PVP, and magnesium stearate were added after the dry ointment was shatter into powder, then the polysaccharide tablets were prepared after the mixture was dried, sifted and tabletted. The polysaccharide was identified successfully by TLC. The linear curve of catalpol concentration within the range of 0.406-2.03 g/1 was ideal (R2=0.9998). The retrieve rate was 96.2%, and RSD= 1.70%.Conclusions:The optimum extraction procedure is stable and reliable, it can be used in the extraction procedure of persimmon leaves flavonoids tablet. The quality control method is accurate, stable and reliable, it can be used in the quality control of persimmon leaves flavonoids tablets. Objective:1. To study the quality of polysaccharide tablet, establish the method for its quality control.Methods:The optimum procedures in two-step process water extracting and alcohol precipitation were selected by the orthogonal text design L9 (34) respectively. The yield of the dry ointment was used as the assessment in the water extracting, and the polysaccharide extract was used as the assessment in alcohol precipitation. The content of polysaccharide was determined by spectrophotometry. The microcrystalline cellulose, povidone iodine, and magnesium stearate were added after the extractive of YTG was shatter into powder, then YTG polysaccharide tablets were prepared after the mixture was dried and sifting. YTG polysaccharide was identified by TLC. The contents of polysaccharide were determined by spectrophotometry.Results:1. The optimum condition was that the crude drugs were decocted for 3 times with 8-fold water (with 9-fold water for the first time),1.5 h each time. The physic liquor extracted by water was filtered and concentrated to 0.7 g·ml-1 (the proportion of crude drugs to physic liquor), and then precipitated by alcohol. The alcohol concentration for precipitation was 75%, and the time for precipitation was 8 h. The physic liquor was centrifugated, concentrated and dried by microwave vacuum concentration dryer to dry powder, and the microcrystalline cellulose, pvp, and magnesium stearate were added after the dry ointment was shatter into powder, then the polysaccharide tablets were prepared after the mixture was dried, sifted and tabletted.2. The polysaccharide was identified successfully by TLC. The linear curve of polysaccharide concentration within the range of 0.406-2.030 g·l-1 was ideal (R2=0.9988). Objective:To observe the effects of YTG polysaccharide tablets on glycemia and lipidemia in diabetic mice induced by streptozotocin (STZ).Methods:The experimental mice were randomly divided into 6 groups: control group, model group, DMBG group, and high, middle and low dosage groups of YTG polysaccharide tablets. Except the control group, typeⅡdiabetic mouse model was established in all groups induced by intraperitoneal injection of STZ (150 mg-kg-1). After the drugs were administrated to experimental mice for 14 days, the fasting blood glucose (FBG) and blood lipid were determined in diabetic mice.Results:Compared with the model group, high dosage group of YTG polysaccharide tablets could significantly decrease blood glucose in STZ-induced diabetic mice and decrease the contents of TG, TC and LDL-C in the serum and increased HDL-C content in the serum.Conclusion:YTG polysaccharide tablets can significantly reduce fasting blood glucose level in diabetic mice induced by STZ, and adjust metabolic disturbance. Chloroform fraction,ethyl acetate fraction, n-butanol fraction and water-extract and ethanol-precipitate fractions are main effective composition of effect. Objective:To study the antioxidative in vitro from YTG polysaccharide tablets. Methods:The blood was collected by pulling the eyeball after the drugs were administrated to experimental mice for 14 days, and centrifuged. Tthe upper serum was taken for detecting activities of content of GSH, NO and NEFA SOD and MDA level in mice liver were both measured according to the methods of assay kits. Results:YTG polysaccharide tablets increased the contents of GSH and decreased the concents of NO and NEFA in serum. They increased the activity of SOD, and decreased the concentration of MDA in mice liver. Conclusion:Extracts from YTG posses a good antioxidative effect and increase immunity.
