Objective To make the animal model of heroin addiction and readdiction by imitating the processes of human heroin addiction and study the phosphorylational Tau,the formation of paired helical filaments(PHF) and the mechanism in the frontal lobe cortex at the viewpoint of cAMP dependent protein kinase(PKA), which will give a reference to the mechanism and treatment of brain damage induced by heroin addiction and readdiction.Methods 30 SD rats were randomly divided into model group and control group, and the model group established the rat model of heroin addiction and readdiction by the processes of human heroin addiction, abstention treatment to heroin readdiction, readdiction, reabstention treatment to heroin readdiction and third readdiction. The model group rats were injected heroin 1.5mg/kg at the first time and increased 1mg/kg in the follow day, which last 10 days. The model group was accepted abstention treatment after appearing withdrawal symptoms proving addiction followed promoting addiction by substitute-degression principle. The model group establishment included 3 addiction and 2 abstention treatment, and the control group was injected equivalent Physiological Saline. We detected the levels of Tau protein phosphorylation and PHF at Ser396 site by Western blot in the frontal lobe cortex of brain tissue after the third addiction, also detected the levels of PKA-α,PKA-βof all experimental samples for primary discussing reasons of above changes at the same time.Results By contrast, the level of Tau protein phosphorylation at Ser396 site was increased,the formation of PHF was also increased correspondingly at the same site and the level of PKA-αand PKA-βwere also increased in model group.Conclusion Tau hyperphosphorylation and PHF were formed in the frontal lobe cortex of brain tissue of heroin addicted and readdicted rats, which was associated with the rise of PKA.
