Objective : To examine the HIF-1αand Glut-1 expression in mucoepidermoid carcinoma of salivary glands(MEC) ; to investigate the relationship of these markers with clinicopathologic features and the corelation of the two; to explore their significance in the diagnose and prognosis of MEC;and thereby find a new treatment target so as to pave a new way for the genetic treatment of MEC.Methods :The samples of MEC are all taken from the pathological department of The First Hospital of Shanxi Medical University from 2000 to 2009,sixty samples altogether, twenty normal salivary glands tissue . Quantitative immunohistochemistry analysis was used to examine HIF-1αand Glut-1 expression in MEC. Envision PV-6000 Power Vision TM is used to stain and the result is analyzed by using some relevant statistical methods,such as SPSS 13.0,χ2 test, Spearman..If P<0.05,the difference is considered as statistical meaning.Results :1. In lesions of 60 MEC patients,HIF-1αis mainly expressed in karyon and small amounts in cytoplasm.The positive expression rate of HIF-1α(65%)in MEC was higher than that in normal salivary glands(0%),which indicates that HIF-1αparticipates in the occurrence and development of MEC.There was no correlation between the expression level of HIF-1αand sex(P=0.251), age(P=0.244),lymph node metastasis(P=0.25).The expression of HIF-1αin MEC tissues correlate with histological differentiation ,tumour size and clinical stage (P<0.05); the positive expression rate of HIF-1αin the high middle and low histological differentiation is 35%,77.8%,81.8%; the positive expression rate of HIF-1αin T1+T2 and T3+T4 is 44.8%,83.9%;the positive expression rate of HIF-1αinⅠ+ⅡandⅢ+Ⅳis 48.5%,85.2%;it can be seen that the lower histological differentiation,the higher malignant level ,the faster speed of tumor growth ,the higher anoxic level lead to the higher expression level of HIF-1α.2.In lesions of 60 MEC patients,Glut-1 is mainly expressed in cell membrane and small amounts in cytoplasm.The positive expression rate of Glut-1(72%)in MEC was higher than that in normal salivary glands(0%),which shows that Glut-1 correlate with the occurrence and development of MEC.There was no correlation between the expression level of Glut-1 and sex(P=0.714), age(P=0.485),lymph node metastasis(P=1.0).The expression of Glut-1in MEC tissues correlate with histological differentiation ,tumour size and clinical stage (P<0.05); the positive expression rate of Glut-1in the high middle and low histological differentiation is 50%,77.8%,86.4%; the positive expression rate of Glut-1 in T1+T2 and T3+T4 is 55.2%,87.1%;the positive expression rate of Glut-1αinⅠ+ⅡandⅢ+Ⅳis 57.6%,88.9%;it can be seen that the higher pathology classify,the bigger tumor tissue ,the higher malignant level ,the faster speed of tumor growth ,the higher anoxic level need the more glycolysis provide energy ,lead to the higher expression level of Glut-1.3.Positive correlation was found between HIF-1αand Glut-1 expression(rs=0.463,P<0.05),which hints that HIF-1αmay regulate the the expression level of Glut-1 to meet the need of glyco-consumption in the process of growth of MEC.Conclusion:The high expression of HIF-1αand Glut-1 were closely associateded with histological differentiation ,tumour size and clinical stage.The aberrant expression of HIF-1αand Glut-1 may provide a useful prognostic marker in MEC.
