Objective:To investigate the effects of rosuvastatin on expression of matrix metalloproteinase-9(MMP-9) mRNA and protein in cultured human monocyte-macrophages cells(HMMC),thatinduced by oxidized low density lipoprotein(ox-LDL).Method:Human monocyte-macrophages cells were incubated to culture 2-4 generations for trail invitro by density gradients centrifugation, and the cells cultured for24 hours as control trial.Theox-LDL control group is incubated for 24 hours by only ox-LDL. Experimental group werecultured with ox-LDL (100 mg/mL) for 2 hours after, and were added to different doses(0.01,0.1,1.0 and10.0μmol/L) of rosuvastatin respectively for 24 hours. Total RNA of everygroup cells were abstracted and MMP-9 mRNA were determined by reversetranscription-polymerase chain reaction(RT-PCR). The enzyme-linked immunosorbentassay(ELISA) were performed to determin the protein contentof MMP-9.Results:(1)The expression of MMP-9 and protein increased significantly forHMMC induced byox-LDL(100 mg/mL), which is in comparison with the control group.(2)The different does of rosuvastatin group can inhibit MMP-9 and protein expressioncombined with ox-LDL control group(P<0.05), and this was in a concentration-dependentfashion.(3)The rosuvastatin10.0μmol/Lgroupof MMP-9 protein expressionwas no significantchanges in comparison with the control group,that was not significant in statisticallyrespect(P>0.05)..Conclusion:The expression of MMP-9mRNA and protein were increased significantly afterin HMMCinduced by the action of ox-LDL. Rosuvastatin is able to down-regulate MMP-9mRNA andprotein expression, and play a role in plaque inflammation and stability, corresponding with adecrease of ox-LDL, whichare related to the rosuvastatindifferentdose.
