| background:Acute coronary syndrome (acute coronary syndrome, ACS) happened close relations with thrombosis, tissue factor (tissue factor, TF) way to start the coagulation system in arterial thrombosis plays an important role. Plaques rupture with platelets and blood coagulation cascade activation is ACS pathological physiological mechanism. Tissue factor as activate exogenous blood coagulation system key factors may through the veins and thrombosis and other role and influence of coronary heart disease occurrence and development.Objective:To investigate the effects and mechanism of Rosuvastatin on Expression of Tissue Factor in cultured Human monocyte-macrophages cells(HMMC),that induced by oxidized low density lipoprotein(ox-LDL).Methods:Human monocyte cells were isolated by density gradients centrifugation in vitro and incubated to culture4generations for induced to macrophages cells with Phorbol ester (160nmol/ml) induced24h for trial. The human monocyte-macroph-ages cells(HMMC) were divided into four groups:ox-LDL group, Losartan group, rosuvastatin group and Losartan with rosuvastatin group. The expression of TFmRNA was assayed by RT-PCR., The enzyme-linked immunosorbent assay (ELISA) were performed to determin the protein content of TF.ResultsrThe rosuvastatin group or Losartan group compared with ox-LDL group the expression of TF decreased and this was in a concentration-dependent fashion(P<0.05). The rosuvastatin with Losartan group compared with ox-LDL group-ã€rosuvastatin groupã€Losartan group the expression of TF mRNA decreased and this was in a concentration-dependent fashion(P<0.05). Conclusions:Rosuvastatin and Losartan are able to down-regulate the expression of TF mRNA and protein in HMMC,Reducing the incidence of acute coronary events,and Rosuvastatin with losartan can increase this effect. |
