Objective:To investigate the role of hyperlipemia in the mechanisms of neuronal injury afterreperfusion, we studied the effects of hyperlipemia on the expression of GFAP and bFGF afterhyperlipemia with cerebral ischemic-reperfusion injury.Methods:166 adult male Sprague-Dawley rats weighing (160-180g) g (provided by Animal Unit ofShanxi Medical University) were randomly divided three groups: cerebral ischemia-reperfusionwith normal diet group (n=52) ; cerebral ischemia-reperfusion with high-fat diet group (n=52).and sham group(n=62). After 2h ischemia, the rats were divided into different groups:reperfusion 1d (n=13), 3d (n=13), 7d (n=13), 14d (n=13). Staining and Immunohistochemistrymethod examined the expression of GFAP and bFGF for six rats in each group . For another sixrats, Western blot measure the content of GFAP and bFGF. For another one rat, it is used by TTC.Sham group is used by Staining and Immunohistochemistry (n=48) , Western blot(n=12) andTTC (n=2). high-fat diet rats were fed on high fat standard forage to make hyperlipemia.6 weekslater, we use biochemical method to observe the change of TC,TG,HDL-L and LDL-L. Ifthere was significant difference between high-fat diet group and normal diet groups, the model ofhyperlima was established. On this basis, we prepare the hyperlipimia model.The cerebralischemia-reperfusion model was established with thread embolish of middle cerebral artery .TTC and the stain of neurological behavior are used to test the success of model.Results:1.Many GFAP and bFGF positive cells distributed in hippocampus CA4 after cerebralischemic-reperfusion injury and Almost were astrocyte. .In sham group, GFAP immunoreativecells were sparsely in hippocampus. In normal diet group, GFAP immunoreative cells wereincreased significantly from 1d after reperfusion. With the increase of I/R time, the expression ofGFAP, reached its peak at I2h/3d, begun to decrease at 7d, and continued to decrease at 14d. Incerebral ischemia-reperfusion with high-fat diet group, the GFAP highest point postponed toI2h/7d. The bFGF highest point didn't postpone. Western blot revealed the same regularities.2.Compared with the expression GFAP and bFGF of normal feed rats at each reperfusiontime point, its expression of high-fat feed rats decrease at the corresponding time points .3.The expression of GFAP and bFGF had similar regularities .The scores of relationbetween GFAP and bFGF was 0.5.Results:1.After erebral ischemic-reperfusion, GFAP and bFGF positive cells almost were astrocyte in hippocampus CA4. It has been reported that GFAP and bFGF were involved injury byastrocyte.2.In the two groups of cerebral ischemia-reperfusion group , GFAP and bFGF showed adynamic expression, and had similar regularities in the time of consistent.Correlation analysisshowed a positive correlation, suggesting that: bFGF increased the expression of GFAP .3.Compared with ischemia reperfusion group , the expression of GFAP and bFGF inhyperlipemia with cerebral ischemia-reperfusion group at the same time was significantlylower .,and it scored significantly higher in neuroethology , suggesting that: hyperlipemiaaggravated cerebral ischemia and reperfusion injury.
