Danlongxingnaopian Prescriptions On Cerebral Ischemia And Reperfusion Of CA1 Nestin, Influence GFAP, BFGF And CyclinD1 Expression

Objective:To explore the expressions of Nestin protein, GFAP protein,bFGF protein and CylinD1protein on Danlongxingnao recipe in rats CA1area with focal cerebral ischemia/reperfusion,it would be found the mechanism of Danlongxingnao recipe after focal cerebral ischemia/reperfusion.Methods:The72rats were randomly divided into sham-operation group, model group,Danlongxingnao recipe group and edaravone group. The Longa method was induced in rats to establish the middle cerebral artery occlusion (MCAO) model.Neurological symptom score were evaluated after3d and7d treatment.HE staining of the cells were used in hippocampus CA1area.Nestin protein and GFAP protein were observed by immunohistochemical method on CA1area after focal cerebral ischemia/reperfusion after3d and7d treatment.bFGF protein and CylinD1protein were observed by immunohistochemical method on CA1area after focal cerebral ischemia/reperfusion after ld,3d and7d treatment.The date presented as means±standard deviation,after finishing the database to Excel with the result of statistical software SPSS15.0ANOVA,LSD,with P<0.05was statistically sianificant;statistical non-linear regression analysis using Tamhane’s T2,P<0.05was statistically sianificant. Results:1The model group in hippocampus CA1area had morphologically changed.The Danlongxingnao recipe group made effects on pretection in hippocampus CA1area of morphological changing.2Compared with sham-operation,neurological symptom score of model group had increased and the result was significantly(P<0.05)after3d and7d treatment.Compared with model group,neurological symptom score of Danlongxingnao recipe group and edaravone treatment group had decreased and the result was significantly(P<0.05) after3d and7d treatment.3Compared with sham-operation,the expressions of average optital density of Nestin protein in model group had increased(P<0.05)after3d and7d treatment;the expressions of average gradation of GFAP protein in model group had decreased(P<0.05)after3d and7d treatment;the expressions of average gradation of bFGF protein in model group had decreased after ld,3d and7d treatment(P<0.05);the expressions of average gradation of CyclinD1protein in model group had decreased after1d,3d and7d treatment(P<0.05).Compared with model group,the expressions of average optital density of Nestin protein in Danlongxingnao recipe group had increased(P<0.05)after3d and7d treatment;the expressions of average gradation of GFAP protein in Danlongxingnao recipe group had decreased(P<0.05)after3d and7d treatment;the expressions of average gradation of bFGF protein in Danlongxingnao recipe group had decreased after1d,3d and7d treatment(P<0.O5);the expressions of average gradation of CyclinDl protein in Danlongxingnao recipe group had decreased after1d,3d and7d treatment(P<0.05).Conclusion:Danlongxingnao recipe can make effects on the expressions of Nestin protein, GFAP protein, bFGF protein and CylinD1protein in hippocampus CA1area after focal cerebral ischemia/reperfusion and it may be associated with promoting the proliferation of neural stem cell to promote the neural stem cell.
