Three cells culture process of the morphological changes observed. Changes of HepG2 transfected by pcDNA-3.1/ pcDNA-3.1-IDO were detected by MTT, FCM, cell adhesion assay and tramswell chamber assay in vitro. HepG2, pcDNA3.1HepG2 and pcDNA3.1IDO-HepG2 cells were studied respectively.Results: Compared with the HepG2 and pcDNA3.1HepG2 cells, pcDNA3.1-IDO-HepG2 cells became longer and had more filopodia. FCM showed that the growth of HepG2, pcDNA3.1HepG cells were blocked in G0 /G1 stage; Attachment assay showed that HepG2, pcDNA3.1HepG cells (0.93±0.18,1.07±0.14 ) adhered more slowly to Matrigel than pcDNA3.1IDO-HepG2 cells(1.179±0.36) (P<0.01). By transwell chamber system, the capability of invasion of pcDNA3.1IDO-HepG2 (269.00±21.60)cells was more significantly increased than HepG2, pcDNA3.1HepG cells(185. 60±15.58, 198.80±12.60).
