Objective:To investigate the intervention of Gan Duqing particles on expression of proteins in rats with endotoxemia and liver injury. Methods:(1)Will SD rats randomly divided into four groups: Normal group,Model group,traditional chinese medicine(TCM) group, traditional chinese medicine (TCM) treatment group,each group are only for 10.(2) Model group:Model group rats were administered with distilled water (0..2ml/10g) by gastric perfusion continue for 7 days, once daily. One hour after the last gastric perfusion,rats were injected with LPS (lOmg/Kg) into its abdominal cavity. During the experiment, all rats feed,ordinary normal drinking water, rats must fast for more than 12 hours before intraperitoneal injection and blood sampling. Three hours after the intraperitoneal injected,rats were injected with 1% pentobarbital sodium (30mg/Kg) into its abdominal cavity.Anesthetic rats were ste-rilized with alcohol on the skin,opened belly and sampled blood form abdominal aorta.All of the blood was stored in 4℃refrigerator. The serum differential protein expression profiling was detected by SELDI-TOF-MS technology; Liver function enzyme index test was detected by automated hematology analyzer. Gramnegative bacteria with LPS detection kit (method) detection of endotoxin concentration.10% formalin fixed liver tissues (1.5cm×1.0cm×0.5cm), HE staining routine pathological section were used to observe the pathological changes of liver tissues under light microscope. (3) TCM group,TCM treatment Group:Adult dosage of TCM is 1.58g/crude drug/Kg/per day. Therefore, the conventional dosage of rats are 10.20g/crude drug/Kg/per day which could be obtained by the equation of body surface area. TCM group rats were administered with Gan Duqing particles 0.2ml/10g by gastric perfusion continue for 7 days, once daily. One hour after the last gastric perfusion, rats were injected with the same dose of distilled water into its abdominal cavity;TCM treatment group rats were administered with Gan Duqing particles 0.2ml/10g by gastric perfusion continue for 7 days, once daily, One hour after the last gastric perfusion, rats were injected with LPS (10mg/Kg) into its abdominal cavity. The rest of the experimental method was the same as the model group's. (4)Normal group:Normal group rats were administered with TCM 0.2ml/10g by gastric perfusion continue for 7 days, once daily. One hour after the last gastric perfusion, rats were injected with the same dose of distilled water into its abdominal cavity.The rest of the experimental method was the same as the model Group's. Results:(1) Each group's serum Endotoxin level:compared with model group, the serum endotoxin level was decreased in TCM Treatment group, the differences were statistical significance (P<0.05).(2) Liver biochemistry test showed:compared with model group, the levels of serum ALT,AST were decreased in TCM treatment group, the differences were statistical significance (P<0.05).(3) 110 differently expressed protein peaks were found by protein chip, among which 15 differently expressed protein peaks had statistical significance (P<0.05). Relative quality than for Hollywood 9193.117 Da protein peak in the normal group and the model group between expre-ssion. Relative quality than for Hollywood 2844.562 Da protein peak in the normal group and the TCM group between expression. Relative qual-ity than for Hollywood 3827.878Da protein peak in the model group and the TCM treatment group between expression.(4) Observed under optical microscope:①Normal group:Normal liver cells and nuclear membrane clear.②Model group:Most of the liver tissue is destroyed, liver cell de-generation necrosis,hemorrhage, without obvious regeneration phenome-non.③The TCM group with the normal group.④the TCM treatment group:will a small amount of the liver tissue damage, stem cells without apparent degene ration necrosis and nuclear membrane is clear. Conclu-sions:(1) The Gan Duqing particles can reduce the serum endotoxin level in rats with endotoxemia and livery. (2) The Gan Duqing particle can reduce the level of ALT,AST in rats with endotoxemia and liver injury. It can promote liver rejuvenesce. (3) The Gan Duqing particles have certain intevention role to expression of proteins, which have an effect on expression of proteins in rats with endotoxemia and liver injury. Change in protein peak,suggesting the differential proteins may be the treatment of Gan Duqing particles molecular target of endotoxemia.The differences in serum protein exists, endotoxemia in the pathogenesis of liver injury and its treatment has a guiding role,for research in the field of molecular evidence.
