Background:PRF can promote proliferation and differentiation of different kinds of cells. From a radiologic and histologic point of view at 6 months after surgery,the use of Choukroun's PRF as the sole filling material during a simultaneous sinus lift and implantation stabilized a high volume of natural regenerated bone in the subsinus cavity up to the tip of the implants.People found that platelet-rich fibrin modulates the expression of extracellular signal-regulated protein kinase and osteoprotegerin in human osteoblasts. Approximately 97% of the platelets and >50% of the leukocytes were concentrated in the PRF clot and showed a specific three-dimensional distribution, depending on the centrifugation forces.α- granular of platelet can release many kinds of cytokines.The leucocytes of PRF can secrete 3 proinflammatory cytokines (IL-1beta, IL-6, and TNF-alpha), an antiinflammatory cytokine (IL-4), and a key growth promoter of angiogenesis (VEGF).Purpose: Study whether PRF can influence the proinflammatory cytokines'release of human gingival fibroblasts.Methods:Culture the primary human gingival fibroblasts,then stimulate the cells to release the proinflammatory cytokines by Pg-LPS. Whether PRF influence the proinflammatory cytokines'release of human gingival fibroblasts were studied by ELISA and Immunocytochemical staining.Results:Pg-LPS which concentration is 0.1ug/ml can stimulate HGF to release IL-6,TNF-α,MCP-1;We get PRF from 3-ml blood samples,and 1/4 of PRF is suitable to our study;PRF can release IL-6,TNF-α,MCP-1 by itself;PRF can weaken the effect of Pg-LPS; PRF participate in the immune response of HGF.Conclusion:Ⅰ:PRF can release IL-6,TNF-α,MCP-1 by itself;Ⅱ:PRF can weaken the effect of Pg-LPS;Ⅲ:PRF participate in the immune response of HGF.
