Background and Objective:Nowadays, implant therapy has been developed rapidly in clinic use. Implant restoration has been accepted as the best way of teeth and dentition defection. With the maturity of the implantation technology, the osseointegration between the implant and the alveolar bone is no longer the success standard of implant restoration. Aesthetic effect has become one of the most important factors for success.Currently, a variety of soft tissue treatment has been used. Not only the surgical method is relatively complex, with high request of the surgeon, but also the postoperative effect is not stable, and make the patient painful by the repeated operations. In 1984, Assoian first prepared platelet-rich plasma(PRP), which was confirmed playing an excellent role in the repairing and regeneration of soft tissue and bone tissue by a large number of laboratory and clinical experiments. However, with the addition of artificial agents, the PRP may cause immune rejections as an antigen. Moreover, the complex production process and the lack of unified standard limits the clinical application of PRP. In 2001, the platelet-rich fibrin(PRF) which was considered as a new generation of platelet concentrates was first discovered by Choukroun. Without any artificial agents, it can avoid immune rejections and cross-infections. PRF is rich in growth factors, and, with the slow and nature polymerization, simulating the physiological fibrin formation, it's similar to the structure of the fibrin in the normal blood clot. This special structure have a strong affinity to a variety of growth factors(GFs), which can make GFs released slowly and extend the treating time of PRF. Recently, more and more literatures suggest that PRF can shorten the healing time and increase the quantity and quality of gingival tissue, PRF may be a new way to improve the aesthetic effect of the soft tissue of implantation.The objective of this study is to observe the human gingival fibroblasts'biological behavior with the use of PRF, which has the special composition and structure, and to analyse the role of PRF in gingival repair and regeneration. With this theoretical guidance, we can promote the use of PRF in clinical application.Methods:In this study, we got the healthy human gingival tissue to perform fibroblast primary culture. Identify the cells by morphological an immun- ological method, then draw the cellular growth curve. We got 2 ml human venous blood to prepare PRF. The blood and cells were harvested from the same patient for immune compatibility reasons. We selected the fibroblasts of 4-7 passages for the culture with PRF, and devided into blank control group (control), one PRF membrane group(P1), two PRF membranes group(P2). Run the MTT assay and ELISA assay of COL-â… and hyaluronic acid(HA) with the supernatants for 1day, 3days, 5days. The data was disposed with statistical software SPSS 17.0.Results:In the primary culture of gingival fibroblasts, we could see the cells long and stringy, with plump cytoplasm, round nucleus and prominent nucleolus. Anti-vimentin staining was positive and anti-keratin staining was negative. The cellular growth curve presents the shape of"slack-exponential growth- platform".
