ObjecttvePlatelet activity and increased arterial stiffness are strongly associated with atherosclerosis.To evaluate atherosclerosis and risk factors in subjects with impaired glueose tolerance(IGT) by measuring of platelet activity markers (CD62P,PAC-1) and brachial-ankle pulse wave velocity (baPWV).To investigate the association between platelet activity and arterial stiffness.Method35 subjects with IGT(group A),35 subjects with type 2 diabetes mellitus (group B),40 normal control (group C)are inculded in the study.To measure ba- PWV by VP-1000. Flow cytometric analyses is performed to measure percentage and mean fluorescence intensity of CD62P and PAC-1.Changes of count of blood platelet,mean platelet volume(MPV), platelet distribution width(PDW) and Platelet-large cell ratio(P-LCR)are analyzed.TG,TC, HDL-C, LDL-C, VLDL-C, FBG are measured simultaneously. To evaluate the risk facto-rs of atherosclerosis in subjects with IGT. Results1.MPV(NC:9.45±0.92,IGT:10.45±0.78,DM:10.59±0.89,P<0.05),PDW(NC: 11.23±1.54,IGT:12.76±1.36,DM:12.91±1.78,P<0.05),P-LCR(NC:0.26±0.051,IGT:0.30±0.063,DM:0.31±0.06,P<0.05)are significantly increased in group A and group B.There are no changes between group A and group B.2.Compared with group C, CD62P(%)(NC:2.34±0.91,IGT:4.02±1.35,DM: 5.25±2.86,P<0.01),CD62PMFI(NC:13.86±5.23,IGT:20.19±5.07,DM: 21.01±6.58,P<0.05),PAC-1(%)(NC:9.45±6.21,IGT:24.80±10.83,DM:25.30±14.42,P<0.01)are significantly increased in group A and group B.They are no significant increased between group A and group B. PAC-1MFI (NC:14.03±5.1,IGT:15.41±3.29,DM:16.86±8.35, P>0.05) is no significant in three groups.3.baPWV(NC:1307.63±160.89,IGT:1470.05±312.04,DM:1604.04± 230.39)is significant increased in three groups.4.In group A, pereentage of CD62P is positively correlations with DBP (r=0.397,P=0.018),SBP(r=0.487,P=0.003),BMI(r=0.437,P=0.015);CD62 PMFI is positively correlations with MPV(r=0.385,P=0.023),PDW (r=0.376,P=0.026),P-LCR (r=0.483,P=0.003);Pereentage of PAC-1 is positively correl-ations with LDL-C(r=0.371,P=0.028),MPV(r=0.406,P=0.016),P-LCR (r=0.346, P=0.0426);PAC-1MFI is positively correlations with MPV (r=0.400,P=0.017),P-LCR(r=0.355,P=0.036).5.In group A,baPWV is positively correlations with SBP(r=0.678,p=0.000),FBG(r=0.349,p=0.017),CD62P(0.394,p=0.019).6. By stepwise regression analysis in group A, P-LCR (b=34.158,P=0.008),LDL-C(b=4.537,P=0.013),TC(b=2.479,P=0.058),BMI(b6=0.276,P=0.026) are contributors to CD62P;P-LCR (b=20.102,P=0.067),MPV(b=4.762,P=0.031)及DBP(b=0.447,P=0.072) are contributors to PAC-1。7.By stepwise regression analysis in group A,FBG(b=102.016,P=0.046) and SBP(b=8.078,P=0.000) are contributors to PAC-1. Conclusions1.There is same degree of platelets activition between objects with diabetes mellitus and impaired glueose tolerance.2.MPV(fl),PDW(%) and P-LCR(%) can also reflect platelets activition in group A.3. BaPWV increases with the increased level of glueose,which demonstrat- es that arterial stiffness increases with the increased level of glueose.4. Increased platelets maybe contact with the increased arterial stiffness. Further study whither platelets activition contribute to the increased of arterial stiffness should be continued5.In group A, LDL-C,TC,DBP and BMI are risk factors of platelets activition.
