| Objective:To explore the association between AFB1 exposure and DNA damage repair mechanism for better understanding of hepatocarcin-ogenesis.Methods:Immunohistochemistry method was used to detect the levels of hOGGl, MGMT and AFB1-DNA adducts. We collected 428 cases of HCC and non-HCC liver tissues were categorized into 7 groups according to their residency, HCC (n=103) and control groups (n=58) from Nanning area, HCC(n=89) and control groups (n=55) from Liuzhou area, HCC (n=86) and control groups (n=37) from Guilin area.Results:In Nanning, Liuzhou and Guilin, the levels and mean optical density of hOGGl protein expression in HCC groups are higher than control groups (P<0.05). The level and mean optical density of hOGGl protein expression in Nanning HCC groups are higher than that of Guilin HCC groups(P<0.05), the similar situation as control groups(P<0.05); In Nanning, Liuzhou and Guilin area, the levels and mean optical density of MGMT expression in HCC groups are abnormal lower than those of control groups(P<0.05), there are no difference among the three HCC groups (P>0.05), the same situation as control groups (P>0.05).Conclusions:There are regional differences of DNA oxidative damage repair and AFB1 exposure in Guangxi, the levels of hOGGl expression in Nanning area are much higher than that of Guilin area. Abnormal expression of MGMT may play an important role in hepatocarcinogenesis. There may be an association between high level oxidative stress and AFB1 exposure, high levels of oxidative stress may be an important mechanism of AFB1 giving rise to HCC. |
