Object:Using polymerase chain reaction and multiplex PCR,utilizing a simple,accurate and economical detection method of metabolic enzyme gene polymorphism.To study the genetic polymorphisms of GSTM1 and GSTT1 in Zhuang people of Guangxi and the relationship between genetic polymorphisms of GSTM1 and GSTT1.Revealing the inner mechanisms of lung cancer.By means of looking forward to early diagnosis and prevention in lung cancer and to provide a theoretical basis.Methods:This study was based on a case-control study.A total of 68 lung cancer pations were selected from in-pations in the Thoracic Tumor Scetion and Chemotherapy of First Affiliated Hospital of Guangxi Medical University.While the control group selected from Respiratory Medicine ,70 cases of patients in the corresponding time period,which were non-neoplastic diseases of respiratory system.Both groups are the Zhuang people in Guangxi .To collect questionnaire study of smoking history,occupational history and so on.Using PCR and muliplex PCR amplification of two groups GSTM1,GSTT1DNA, gel electrophoresis system to determine genotype,DNA repeat genotypes verify the accuracy of detection. Using chi-square test, t test and logistic Regression statistical methods to analyze the expression of GSTM1, GSTT1 in lung cancer population and control groups with the relationship between clinical indicators, including age, gender, clinical stage and pathological types; To analyze the combined effects in the lung cancer by combination the factors of Smoking and GSTM1, GSTT1 genotype polymorphism. Results:(1)The frequencies of GSTM1(-) and GSTT1(-) in the non-lung cancer people of the Zhuang people in Guangxi are 55.71% and 48.57%;(2)GSTM1 genotypes were consist of function and defective,defective is for the complete absence of GSTM1 gene(GSTM1-null),the frequencies of GSTM1(-) in lung cancer group and control group were 69.12%(47/68) and 55.71%(39/70),chi-square value is 3.105,the difference was not significant(P=0.078);(3) GSTT1 genotypes were consist of function and defective,defective is for the complete absence of GSTT1 gene(GSTT1-null),the frequencies of GSTT1(-) in lung cancer group and control group were 66.18%(45/68)and 48.57%(34/70),chi-square value is 2.330,the difference was not significant(P=0.127);(4) We used the analysis of GSTM1 combine with GSTT1 and found that the frequencies of GSTM1(-) and GSTT1(-) in the cancer group and control group are 50.0%(34/68) and 27.14%(19/68), it's showed that combination of two defective genotpyes was strongly associated with lung cancer, chi-square value is 5.185(P=0.034),carring GSTM1(-),GSTT1(-) genotype risk of lung cancer is to carry GSTM1(+),GSTT1(+) genotype of 3.102 times(95%CI1.230~7.806);(5) By analysis of GSTM1 combine with smoking status, to suppose nonsmokers carried GSTM1(+) OR=1,then GSTM1(+) with smoking individuals increased lung cancer risk of 1.912 times,nonsmoking population,GSTM1(-) had a 2.214 fold increased risk of lung cancer compared with those GSTM1(+);smokers with GSTM1(-) the OR was increased to 3.778 times(95%CI为1.890~12.194,P=0.026);(6) By analysis of GSTT1 combine with smoking status, to suppose nonsmokers carried GSTT1(+) OR=1,then GSTT1(+) with smoking individuals increased lung cancer risk of 1.481 times,nonsmoking population,GSTT1(-) had a 1.667 fold increased risk of lung cancer compared with those GSTT1(+);smokers with GSTM1(-) the OR was increased to 2.883 times(95%CI为1.170~8.340,P=0.061);(7) The distribution of frequencies of GSTM1,GSTT1 genotype in lung cancer clinical stage and histological type showed no significant difference(P>0.05). Conclusion:(1) There was no significant difference of the frequencies of GSTM1,GSTT1 genotpyes between the Zhuang people and the healthy Han people in Guangxi while comparing with the documents.(2) GSTM1,GSTT1 do not increase the risk of lung cancer when analyze its single genotype defective in Zhuang people of Guangxi ,while combination of two genetpyes was significant associated with lung cancer susceptibility,carrying GSTM1(-),GSTT1(-) individuals susceptible to lung cancer.(3)Smoking and GSTM1,GSTT1 gene polymorphisms have synergistic. Smoking and GSTM1-null genetype seemed to be increase the risk of lung cancer.
