Association Of CYP1A1 And XRCC1 And GSTM1 Polymorphisms With Genetic Susceptibility To Lung Cancer

ObjectiveTo investigate the relationship between the susceptibility to lung cancer and gene polymorphism of cytochrome P450(CYP1A1) and base excision repair gene (XRCC1) and Glutathione-S-transferase M1(GSTM1).MethodsThe polymorphisms of cytochrome P45O(CYP1A1) and base excision repair gene XRCC1and Glutathione-S-transferase M1(GSTM1) were detected by AS-PCR and PCR-RFLP methods among 209 lung cancer cases and 208 healthly controls who had physical examination in First Affiliated Hospital of Zhengzhou University.A case-control study was used on association between CYP1A1, XRCC1, GSTM1 and lung cancer susceptibilityResultsFrequencies of CYP1A1 Val/Val genotype were12.1% in control and 16.7% in cases, respectively, and there were significant difference (P=0.041). Compared with Ile/Ile genotype,carrying both Ile/Val and Val/Val genotype increased the risk in squamous and adenocarcinoma cell cacinoma and there was statistical significance in the risk of squamous cell carcinoma (OR=1.161,95%CI=1.036-1.302,P=0.018). The interaction of smoking and Val genotype was observed (I(AB)=2.155).Frequencies of XRCCl Trp/Trp genotype cases(14.3%) was significantly lower than controls(7.6%) (P=0.024).Compared XRCCl Trp/Trp with XRCCl Arg/Arg genotype, the risk in squamous and adenocarcinoma cell carcinoma with Trp genotype increased. Trp genotype increased risk in squamous cell carcinoma (OR=1.577,95%CI=1.143-2.176,P=0.005).In comparison with Arg/Arg genotype, there wasinteraction between smoking and Trp mutant genotype (I(AB)=4.349) There were significant difference between frequency of GSTMl(-) genotype patients(58.4%) and controls(42.8%)(P=0.001).Compared with GSTM1(+) genotype, GSTMl(-) genotype increased the risk in squamous and adenocarcinoma cell carcinoma.And GSTMl(-) genotype was statistically significant risk factor for squamous cell carcinoma (OR=2.106,95%CI=1.300-3.411, P=0.002); A positive interaction was found between smoking and GSTMl(-) mutant genotype (I(AB)=2.505). A positive interaction was found between Val genotype and GSTMl(-) mutant genotype (I(AB)=1.536); A positive interaction was found between Trp genotype and GSTMl(-) genotype(I(AB)=2.824); A positive interaction was found between Tip genotype and Val genotype (I(AB)= 1.515);ConclusionSusceptibility to lung cancer was ehanced by the presence of genetype CYPlAl Val, XRCC1 194Trp, GSTMl(-). The genotyps of CYPlAl Val, XRCC1 194Trp, GSTMl(-) might be the risk genotypes for squamous cell carcinoma and adenocarcinoma, but only the inceased risk in squamous cell carcinoma was statistically significant difference. There was significant interaction between the genotype of CYPlAlVal, XRCC1 194Trp, GSTMl(-) and smoke in susceptibility to lung cancer...
