objective: Epilepsy is a common chronic disease of the nervous system, genetic factors are one of the factors of its causes. To identify and analyze the leading cause of differences between different groups of people , human population genetics studies often Comparise the presence of a particular trait among the genes. Gene research can ultimately found that the responsibility gene (or genome), especially when we cannot obtain biological information to explain the origin of the disease. so successful genetics research can broaden the idea of the researcher ,which is possible to discover the related factors of the process of this disease, which is completely unknown in the past or not suspected. This experiment is to research the relationship between 5-HTR2A expression change in frontal lobe and temporal lobe brain tissue of epilepsy patients and pathogenesis of epilepsy, which can provide a theoretical basis for clinical diagnosis ,treatment and prevention. Methods : The brain tissues in experimental group are from epilepsy lesions resection patients in chengdu 363 hospital. Before the operation, all patients must be monitored by long video EEG, epileptic focus is clearly located in the frontal and temporal lobe by dipole localization, These patients and the family members have annotated know all. According to epilepsy's etiology classification, all patients are in accordance with idiopathic epilepsy. There are no obvious cause, no focal positive signs, no positive imaging studies . The course of the patients is more than 2 years, the curative effect is not good. Among them there are 13 patients of the frontal lobe, with an average age of 21.77 years old; there are 22 patients of temporal lobe, with an average age of 24.75 years old.The brain tissues incontrol group is from Neurosurgery patients with intracranial decompre ssion in chengdu 363 hospital. Among them there are 11 patients of the frontal lobe, with an average age of 51.45 years old; there are 3 patients of Temporal lobe, with an average age of 27.33 years old. This experiment is using real time PCR to detect 5-HTR2A expression change between frontal lobe and temporal lobe brain tissue of epilepsy patients and non-epilepsy patients and using independent samples t test to compare the differences between the two groups. Results: The average expression of 5-HTR2A in frontal lobe brain tissue of epilepsy patients and non-epilepsy patients is 8.72±3.75 and 3.71±2.14 respectively. The average expression of 5-HTR2A in frontal lobe brain tissue of epilepsy patients is 2.35 times of non-epilepsy patients. There had obvious difference between the two groups (P<0.05). The average expression of 5-HTR2A in temporal lobe brain tissue of epilepsy patients and non-epilepsy patients is 0.132±0.073 and 0.121±0.024 respectively. There had no obvious difference between the two groups (P > 0.05). Conclusions: (1) the average expression of 5HTR2A of frontal lobe is more than that of temporal lobe. in physiological conditions. (2) The expression of 5-HTR2A in frontal lobe brain tissue of epilepsy patients is obviously higher than non-epilepsy patients,that is to say 5-HTR2A gene may increase the incidence of epilepsy and in the central nervous system may have the role of enhancing cell excitability, The increase of 5-HTR- 2A expression may be one of the factors of epilepsy.
