Cancer is one of the serious illness that endangering human life and health. How to prevent and treat cancer is still a major challenge facing humanity In the 21 st century. Research and development of anticancer drugs has become an important topic in recent years. Process of tumor invasion and metastasis is the process tumor cells from primary tumor to the surrounding or distant organization. Involved in tumor cells through the extracellular matrix barrier, vascular wall basement membrane and the process of piercing vessel wall into the host micro-environment. A large number of experiments to prove that ability of tumor cell invasion and metastasis is closely related to the ability that induced to generate protease to degradation of extracellular matrix and GBM. MMP can degrade virtually all ECM components, which is the most relevant enzymes with tumor invasion and metastasis. MMPs are ECM proteolytic enzymes, because the diversity of ECM structural components and complexity of the cell role, MMPs degrade ECM proteins at the same time, inevitably affect the cell signal transduction and cell differentiation, morphology and function.so, MMPs through degradation of ECM components to mediate its dynamic balance between the recombinant protein and the degradation substrate. In order to eliminate of certain special signals, show implicit signal, so that make already exists in the matrix of biological active substances released or activated, greater involvement of cells and tissues and organs of various forms and functional changes. A MMP can directly degrade a certain one or a few ECM, could also play the role by activating other types of MMPs. MMPs mainly have three kinds of mechanisms to promote the invasion and growth of tumor cells:①The role of protease makes physical barrier around the tumor cells destroyed;②MMPs can enhance the cell adhesion, so that the growth of tumor cells to the surrounding;③after MMPs acting on the matrix components, inspired a number of other potential biological activity.Inhibitor of MMPs include non-specific inhibitor, TIMPs and synthesis inhibitors. Which TIMPs are specific inhibitors of MMPs, in regulating the activity of MMPs plays a most important role in. TIMPs are endogenous tissue-specific inhibitor which secreted by the body cells, can inhibit the activity of MMPs, and can combine with MMPs'plasminogen in activated form. TIMPs in the tumor cells and stromal cells can be expressed. The primary function of TIMPs is that inhibiting MMP activity, preventing MMP-mediated endothelial cell movement, inhibiting the release of vascular endothelial growth factor in substrate for inhibiting angiogenesis, preventing the degradation of extracellular matrix. Therefore, whether from a traditional Chinese medicine to develop new inhibitors of MMPs is a new breakthrough in. Extracted antitumor drugs from natural plant is currently an important part of Modernization of Chinese medicine in China, but also foreign-based clinical research hotspot. A number of synthetic anti-cancer chemotherapy drugs, their development of expensive, toxic side effects, and even their own carcinogenic toxic, caused by the occurrence of second tumors; the Chinese herbal medicine, herbal and other natural product exist a wide range of bio-active substances. Traditional Chinese Medicine Treatment of dialectic, the overall principle of treatment is very effective in cancer treatment, so people turned to the natural world. From natural products to look for small toxic side-effects, the role of a unique anticancer drug and auxiliary anti-tumor drugs. A large number of epidemiological investigations confirmed many Chinese herbal medicines had played an important role in cancer prevention and treatment. These natural plant that have anti-tumor and regulate the immune function is expected to become an important source of anticancer drugs. There are broad prospects of development and application value. Chinese herbal medicine in particular has wide variety of sources, a wide variety. According to incomplete statistics, anti-cancer agents from herbal account for 32.25 percent, such yew drunk, camptothecin, vincristine, as a drug of choice for treatment of certain tumors has been widely accepted at home and abroad.The pharmacological effects research of Rhizoma Paridi has confirmed that it has a clear anti-tumor effect and its extracted a variety of chemical constituents has a clear anti-tumor activity, so, The author wants through this experiment to confirm whether the Rhizoma Paridi treat tumor is related to inhibiting the MMPs'activeness.Objectives:To observe whether the extract of the Rhizoma Paridi can inhibit the matrix metalloproteinases, which offer essential theoretical and experimental foundation for the mechanism of the Rhizoma Paridi inhibiting tumor.Methods:Put a mount of MMP-13 solution to the enzyme buffer solution and the Rhizoma Paridi solution of 5mg/mL,4mg/mL,2mg/mL,1mg/mL,0.5 mg/mL and 0.25mg/mL differently, incubate 30 minutes, then add 0.2μg DQ-gelatin and test the substrate. The fluorescence intensity can response the ability of enzyme degrading the substrate.Principles:The fluorescence can be emitted when the enzyme degrades the substrate, therefore testing the fluorescence intensity can response the ability of enzyme degrading the substrate. DQ-gelatin which is signed by the fluorophore is used in the experiment. After the enzyme degrades the substrate, the fluorophore is separated with quenching group. And to some extent, fluorescence intensity is proportional to the speed of enzyme degrading substrate.Results:The extract of the Rhizoma Paridi have the effect of inhibiting MMP-16 and the function depends on the density. That is to say, the Rhizoma Paridi extract on the inhibition of MMP-16 enhances with the increase in drug concentration, which confirms the new drugs extracted from the natural and targeted to MMPs/TIMPs has bright prospects further.Conclusion:The extract of the hempseed can inhibit the activity of the MMP-16, and this kind of function depends on the density.
