Bronchopneumonia is a major cause threatening the health even the life of children because of the high morbidity and mortality. Bronchial pneumonia is caused by bacteria or viruses, in developed countries the main pathogen is the virus, but in developing countries is bacteria. It usually depends on the blood culture to detect. But it needs a few days to analyze the results and the positive rates are low. It can be used the early detection of bacteria. This main topic states using PCR methods to test 16S rRNA gene in blood stream infection with children who have bronchial pneumonia compared with blood culture methods, CRP and WBC Methodology methods.Gene primers are selected from 16S rRNA's conserved region, which extracted DNA to develop polymerase chain reaction (polymerase chain reaction, PCR). The results shows PCR amplification when DNA was extracted from fever children in the same time, it can help to diagnose the children with bronchial pneumonia. Furthermore, blood culture, CRP and WBC can also be used with SPSS 17.0 FOR Windows to analyze data and to test children with bronchial pneumonia.All the PCR is amplified for the 1300bp DNA of bacteria, viruses and human genetic DNA without cross-reaction.In clinical manifestation,48 cases are tested positive in 16S rRNA gene and 12 cases are tested positive in blood culture of the 56 cases of bronchial pneumonia. The studies shows PCR can detect 16S rRNA and its sensitivity, specificity, diagnostic efficiency and diagnostic indicators are:85.71%,94.11%,88.89%,179.82. The high rates of positive PCR in blood culture are significantly different compared with CRP and WBC. So can be used as diagnostic indicators of bacterial infection.16S rRNA PCR method for detection of bacteria can rapidly determine the presence of bacteria in blood samples. The bacterial infection will have high sensitivity and specificityi n the early stage of children's bronchial pneumonia. 16S rRNA gene can detect bacterial infection rapidly to the children. To help early diagnosis, early treatment, early withdrawal and guide clinical medication. The clinical indicationis greater than the other methods, which has good specificity and can be used as diagnostic indicators of bacterial infection.
