Hypertension is the most common cardiovascular disease, often accompanied by fat and carbohydrate metabolism disorders and heart, brain, kidney and retina and other organs with functional or organic changes to organ remodeling characterized by systemic disease with a yearly growing morbidity. In recent years, the researches on hypertension mainly focus on renin-angiotensin-aldosterone system (RAS), in which angiotensin-transferase (ACE) is the most important rate-limiting enzyme RAS activity, which can inhibit ACE activity to release the blood pressure.In present study, the active components from traditional Chinese medicines were investigated using ACE as the target to exploit new drugs of lowering blood pressure. The main contents were as follows:(1) Screening of TCM with ACE inhibitory activity:The inhibitory activity of ethanol extract from different TCM was detected by HPLC method. The results showed that different TCM has different ACE inhibitory activity. Among them, the Citrus medica L showed the highest activity.(2) Purification of active fractions in Citrus medica L:Citrus medica L was extracted with different solvents and purified by gradient elution, and then the ACE inhibitory activity of various fractions was measured by HPLC. The results indicated that:methanol extract with the same concentration showed the highest activity among different solvent extracts. And the fractions purified by gradient elution showed an improved activity, especially, the fraction eluted by 40% methanol had the strongest inhibitory activity which showed a dose-dependent relationship.(3) Animal experiment:according to the experimental results in vitro, the lowering blood-pressure effect of fraction with the highest inhibitory activity was tested used mice as the model. The results indicated that the fraction can lower the blood pressure in spontaneously hypertensive rats with a dose-dependent relationship.
