On the basis of the theories about senility of tranditional Chinese medicine and modern medicine, the anti-aging effects and mechanism of Jinkui Shenqiwan on aging model mice were investigated.Methods:(1) Aging model mice preparation and treatment:Mice in model control group and test group were subcutaneously injected with D-galactose at the dosage of 100mg/kg per day for 42 days. The mice in test group were intragastricly administered with Jinkui Shenqiwan at the dosage of 1.17g/kg per day for 35 days. The same volume of normal saline was administered to mice in the normal control group.(2) Evalution of aging model:The general physical sign, body weight and serum total antioxidant capacity (T-AOC) of mice in erery group were surveyed.(3) TE contents detection:ELISA was used to detect the TE contents in brain, liver and spleen tissue samples of mice in erery groupe.(4) Cloning of TERT and (3-actin gene fragment:Brain, liver and spleen tissue samples were taken from all mice and the total RNA were extracted. After reverse transcription, TERT andβ-actin gene fragments were amplified from the cDNA with PCR. The gene fragments were purified and cloned into PEZ-T vector, and then sequenced.(5) Detection of the relative expression level of TERT gene:The target gene fragments cloned in PEZ-T vector were used to optimize real-time PCR condition, analyze amplification specificity and efficency and to set up standard curve of the reaction. The relative expression level of TERT gene of mice in every group was detected by real-time fluorescence quantitative PCR using cDNA samples of brain, liver and spleen tissue.Result:The result showed that after Jinkui Shenqiwan administration, the TE contents in brain and liver tissue of aging model mice increased significantly (P<0.01), while that in spleen tissue did not (P>0.05). The relative expression level of TERT gene in brain, liver and spleen tissue increased significantly (P<0.01).Conclusion:This study indicated that Jinkui Shenqiwan could significantly increase the TE contents at protein level and elevate TERT gene expression at transcription level, which might explain one of its anti-aging mechanisms. However, there are so many ingredients in the decoction of Jinkui Shenqiwan, further studies are still needed to analyze the individual active ingredients and their pharmacological effects about anti-aging fuction of Jinkui Shenqiwan.
