Cervical cancer is a higher incidence of female genital malignant tumor, its occurrence, development is a very complexity process of the patho -logical ,and its many factors and many genes that working together. In discussing its occurrence and development process, molecular biology research provides a more reliable basis, that more than 89% cerical cancer human were infected with human papillomavirus (HPV). In addition, tumor-suppressor gene inactivation and hostoncogene activation, certain growth factor, cytokines and its receptor also have a certain stimulative effect with the formation and development of cerical cancer . Invasion and metastasis is an important biological characteristics of malignant tumor, is the main cause of leading death in malignant tumors, its seriously influences tumor prognosis. According to the statistics about 70 percent of cancer patients died of tumor metastasis [1], so how to control and prevention of malignant tumor metastasis are the key to improve tumor patient survival.In recent years, studies have found that ezrin protein (Ezrin) plays a very important role [2] in tumor invasion and metastasis. Ezrin protein is found in chickens intestinal epithelium cells in the microvilli, by Bretscher.Ezrin protion is a member of the ERM protein families.It is a kind of cytoskeleton connected, has 585 protein, amino acid residues, composition, molecular weight is about 82ku, located in chromosome 6q25 - q26, basically participate in epithelial cell membrane dynamics, concentrate on the rich actin cells microvilli and cell membranes. The expression of ezrin can promote tumor metastasis and infestations, and highly correlated different sources of multiple tumors metastasis.A study reported that the expression of ezrin is not only the tumor metastasis from sufficient condition, but also a necessary condition [3]. Ezrin connected the transmembrane cell adhesion molecule is an important physiological function, through participating in regulating cell adhesion molecule of signal transduction pathways, and further promoted the interactions of cell and cellular matrix, cells and cellular. between the variety of cells, and pass with the combination of adhesion molecules, further enhance the adhesion strength, and ligand (extracellular matrix) - adhesion molecules essien this protein - cytoskeleton way also participated in the cells of signal transduction and cell motility [4], adjust the cytoskeleton assembly shrinkage and the expression of cell gene.CD44 is research comparatively mature of the cell adhesion molecule family, its closely linked to cancer occurs, the development, the invasion and metastasis. According to the different explicit expression of ways, CD44 has divided into the son (CD44v) and standard (CD44s). CD44v has a rich expression in tumor, mainly distributed in the epithelial cells. CD44v including three subtypes: CD44v3, CD44v5, CD44v6. CD44v6 is a membrane glycol protein acrossing the cell surface, main function is connected with the cytoskeleton proteins on cells, participated in the formation of locomotion, and related the cell migration sports; can identify and combining with proper ligand, participate in the adhesion between cells and substrate, cells and the cells.As a cells matrix hyaluronic acid receptors, its can change the function and composition of the tumor cell surface adhesion molecule, beneficial to the tumor to transfer and diffusion [5]. The occurrence, development ,invaded and metastasis are associated with the CD44v6 expression in many kinds of human malignant tumor.Objective: The specific mechanism shift of CD44v6 and Ezrin in tumors occurrence and metastasis is unclear.Studies show that ezrin can directly or indirectly combine with CD44v6, further to adjust the tumor cells adhesion , influences the metastasis and metastatic of tumor [6],and the function of ezrin can also be activated by the expression of CD44v6, thereby strengthening transfer ability of the tumor. This study examined the expression of ezrin and CD44v6 in cervical cancer tissue , and discussed the relationship between ezrin ,CD44v6 work together with cervical lymph node metastasis.Methods: Select 95 cases of cervical tissue samples from the First Hospital of Hebei Medical University, Third Hospital of Hebei Medical University, The Second Hospital of Shijiazhuang and the first Central Hospital of Baoding ,within 2005-2010. All the cases confirmed by pathological examination . A total of 95 organizations which Was seized were divided into three groups, A group, 35 cases of cervical cancer, including lymph node metastasis (A1 group) 25 cases ,without lymph node metastasis (A2 group) 10 cases; B group, cervical intraepithelial neoplasia (cervical intraepithelial neoplasia, CIN) tissues of 30 cases; C group of normal cervical tissue or tissue associated with 30 cases of chronic cervicitis. To use Immunohistochemical method to detect organization which were seized. All the statistical analysis complete using SPSS13.0, calculation positive rate ,between the rate usingχ~2 inspection and continuous correctionχ~2 inspection, and analysis thelinear correlation.Results:1 Ezrin protein along the linear distribution, inside the cell membrane and the cytoplasm showed brown particles distribution. In the cervical cells or cervical intraepithelial neoplasia cells ,we can see the Ezrin protein positive signal, but in normal cervical tissue Ezrin protein was not found expression. These results suggest that, the positive expression rate of Ezrin protein in A, B and C group was 65.7%,30.0%,0.0%,compared with three groups, the difference was statistically significant(P=0.000<0.05);three groups pairwise comparison, the differences were statistically significant (P <0.0125). In A1 group , the expression rate of Ezrin protein was 80.0%, In A2 groups , the expression rate of Ezrin protein was 30.0%, the difference was statistically significant (P=0.015 <0. 05).2 The distribution of CD44v6 along the inner membrane, in the cytoplasm or the cell membrane was brown granular performance. The positive signal in the membrane and in the cytoplasm of cervical cells, cervical intraepithelial neoplasia cells or normal cervical tissue . These results suggest that, the positive expression rate of CD44v6 in A, B andC group was 82.9%, 46.7%, 10.0%, compared with three groups, the difference was statistically significant (P =0.000<0.05); three groups pairwise comparison, the differences were statistically significant (P <0.0125). In A1 group , the expression rate of CD44v6 was 96.0%, In A2 groups , the expression rate of CD44v6 was 50.0%, the difference was statistically significant (P=0.004 <0. 05).3 The expression of Ezrin and CD44v6 in cervical cancer has positively related, the correlation coefficient (r = 0.739, P <0.01)Conclusion:1 In the tissues of carcinoma, the posive rates of the expression of Ezrin protein and CD44v6 in the carcinoma tissueswere both higher than their atypicalhyperp lasia of tumor - adjacent and normal cervicalmucosa, there was a statistic correlation,the difference was alll significant between them ( P < 0. 05).2 In matastatic group, the positive rates of expression of Ezrin protein and CD44v6 were higher than those in non– metastatic group. There was a significant difference between the two groups( P < 0. 05).3 The expression of Ezrin protein was positively correlated to that of CD44v6 in cervical cancer ( P < 0. 01) .4 The over expression of Ezrin protein and CD44v6, incidence the development and progression of cervical cancer, and in metastasis cervical cancer playing an important role .The combine promoting of the two protions ,may be becoming an important indicator for metastasis and prognosis of cervical cancer.
