Purpose:To preliminarily investigate the radioprotective effects of the aqueous extract of S.obval-lata DC.Edgew on the hematopoietic system, immune system and sperm of mice from radiation injury.Materials and methods:After intragastric administration of aqueous extract of S. obval-lata DC. Edgew at different doses for 14 days, mice were irradiated with 4 Gy of x-ray, Then the intragastric administration of drugs for low, middle and high dose group were followed on. At different time after irradiation, mice were sacrificed and analyzed with several methods.1. Micronucleus test:1 day after irradiation, mice were sacrificed and measured micronucleus.2. Analysis in the blood system:The mice were killed at 1,3,6,9 and 12 days after the first irradiation, and RBC, WBC, PLTand Hb content from venous blood were measured. The number of mouse bone marrow nucleated cells and bone marrow DNA content (A value) were measured at 3,6 and 9 days after irradiation.3. Analysis in the immune system:The mice body weight, spleen and thymus weight were measured at 1,3,6 and 9 days after the first irradiation.and the spleen and thymus index were calculated.4. Analysis in the reproductive system:35 days after irradiation, mice were sacrificed and the sperm deformity rate were measured. Results:1. Micronucleus test on mice showed that the number of micronuclei in irradiated group was less than positive group (CTX group), but the middle and high dose group was significantly lower than positive group(P<0.01).2. Hematological results showed that WBC, PLT in each treatment group with irradiation were significantly higher than those is irradiated group (P<0.01). And compared with the irradiated group, RBC, Hb content, bone marrow nucleated cell count and bone marrow DNA content (A value) values in the treatment group with irradiation were increased slightly, but there weew no significant statistical difference (P> 0.05).3. Test of immune organ weight and organ index showed that the spleen weight and index values in in each treatment group with irradiation were higher than the irradiated group. Especially, the high dose group was higher than the irradiated group (P<0.05). And compared with the irradiated group, the thymus weight and index values in the treatment group with irradiation were increased slightly, there were statistical difference(P<0.05).4. Compared with the irradiated group, the sperm deformity rates in each treatment group with irradiation were decreased. Especially, the middle and high dose group was significantly lower than irradiated group (P<0.01).Conclusions:The active ingredient of natural herb S.obval-lata DC. Edgew have some radioprotective effects on the blood system, the immune system and the reproductive system in mice.1. In hematopoietic system of mice, the radioprotective effects of S.obval-lata DC. Edgew on WBC, PLT are significant, and the effects on the RBC, Hb content, bone marrow nucleated cell count and bone marrow DNA content (A value) are not obvious.2. In the Immune system of mice, the spleen and the thymus are affected by S.obval-lata DC. Edgew.3. The natural herb S.obval-lata DC. Edgew active ingredient have some radioprotective effects on the germ cells in mice. 4. Micronucleus test on mice showed that the number of micronuclei in irradiated group was less than positive group (CTX group), but the middle and high dose group was significantly lower than positive group. The natural herb S.obval-lata DC. Edgew active ingredient have some radioprotective effects on the chromosome in mice.
