[Objective] To study the BEAS-2B human bronchial epithelial cells stimulated by Nano-quartz, the variations of the cells'morphology, expression of reactive oxygen species(ROS) and cyclin-D1, cell cycle and migration capacity in different time. Explore the Influences of Nano-quartz upon the BEAS-2B human bronchial epithelial cells, laid a theoretical foundation for the further study about the relationship between Nano-quartz and lung cancer.[Method] The experimental group was been cultured by LHC-8 cultivation liquid which contained Nano-quartz 50mg/L, the positive control group was been cultured by LHC-8 cultivation liquid which contained micro -quartz 50mg/L, the control group was been cultured by normal LHC-8 cultivation liquid. When the cells were been cultured 4h,24h,72h, using inverted phase contrast microscope, scanning electron microscope(SEM), image analysis software to analyze the cells' morphology, growth tendency; using flow cytometry (FCM) to detect the cells'expression of ROS,Cyclin-D1 and the variation of cell cycle; Through the transwell chamber migration experiment to detect the change of the cells'migration capacity. Using the SAS 8.0 statistical package to statistical analyses the experiment data.[Results] 1. The variations of the cells'morphology by the extension of culture time, the BEAS-2B cells of the experimental group gradually appearance scale-up, mass of barbs can be seen on the cells'edge, vesicles emersion on the surface, pseudopod formation and finally presented like fungiform bodies. The cells' morphology parameters trending upwards in both experimental group and positive control group, but the trends of the experimental group are more obviously(P<0.01); in the control group, the cells'morphology parameters changes have no statistical significance(P>0.05).2. The expression of ROS By the extension of culture time, the average ROS expressivity of both experimental group and positive control group step-up gradually, and the upward trend of the experimental group are more obviously(P<0.01); the control group's ROS expressivity have no obviously change. 3. Cyclin-D1 By the extension of culture time, the average Cyclin-Dl expressivity, Cyclin-D1 expression rate and the G1 period Cyclin-D1 expression rate of both experimental group and positive control group step-up gradually, and the upward trends of the experimental group are more obviously(P<0.01); the control group's 3 foregoing indexes have no obviously change(P>0.05).4. The variations of the cell cycle The experimental group, positive control group and control group's proportion of G1 period cells have no statistical difference after cultured 4 hours, but by the extension of culture time, the experimental group and positive control group's proportion of G1 period cells step-down gradually, and the decent rate of the experimental group are more obviously (P<0.01), the control group's proportion of G1 period cells have no significant deviation (P>0.05). After cultured 4 hours, the experimental group's proportion of S period cells step-up to 19.7±2.1%, it has statistical significance (P<0.05) when compared with the positive control group and control group. By the extension of culture time, the experimental group and positive control group's proportion of S period cells step-up gradually, and the upward trends of the experimental group are more obviously (P<0.01); the control group's proportion of S period cells have no significant deviation (P>0.05).5. The change of cells'migration capacity After cultured 72 hours, the migration capacity of the experimental group is higher than positive control group and control group, and the result have remarkable statistical significance (P<0.01).[Conclusion] By the stimulation of Nano-quartz, the BEAS-2B human bronchial epithelial cells lose its original morphous, its appearance scale-up, mass of barbs and vesicles can been seen on the surface, pseudopods were formed; The ROS and Cyclin-D1 were overexpression; The cell cycle is transferred into S period from G1 period; The cells' migration capacity is increased; and it shows particle size-effect and time- effect relation.
