ObjectiveDetection of the safety of BCR-ABL-pIRES-GM-CSF fusion gene DNA vaccine against Chronic myeloid leukemia.MethodsSPF-class male BALB/c mice were randomly divided into control group, pIREs group, pIREs-GM-CSF group, BCR-ABL-pIRES group, BCR-ABL-pIRES-GM-CSF group. First, expression of DNA vaccine in muscle at injection region was detected by RT-PCR. Second, expression change of IL-1β, IL-6, TNF-α, INF-γin serum were detected by Western blotting and ELISA assay methods. Finally, the morphology changes of the liver, spleen, kidney and muscle in injection region were observed by optical microscopy method and the subcellular morphological changes of liver and kidney were observed by electron microscopy method.Results1. Generally condition of the normal control and experimental groups were no significant difference observed. The normal control group and experimental groups of body weight were not statistically significant (p>0.05).2. RT-PCR detection showed:BCR/ABL gene was successfully amplified from the muscle in injection region in pIREs group, pIREs-GM-CSF group, BCR-ABL-pIRES group, BCR-ABL-pIRES-GM-CSF group, Showed that the expression of DNA vaccines in vivo.3. Western blotting method and ELISA assay showed:The expression of IL-1β, IL-6, TNF-α, INF-γin serum in control group have no statistically significant compared with experimental groups (p>0.05).4. Optical microscopy showed that the liver, kidney, spleen, and muscle in injection region were no obvious pathological changes in control and experimental groups. Electron microscope showed that the nuclear membrane of liver and kidney in the normal control group and experimental groups were clearly, structure of mitochondria, endoplasmic reticulum and lysosomal were complete,ConclusionBCR-ABL-pIRES-GM-CSF fusion gene DNA vaccine in mice is tolerable and has a good safety.
