| The children anorexia is a kind of feeding disorder in young children. The children suffering from anorexia have a poor appetite or lose their appetite, or even hate to eat their food. The therapy of invigorating spleen is an important theorem, which regulates the function of digestive system and aims specifically at the characteristic of children digestive disorder. The pediatric herbalists have practiced the theorem in clinic for many years and have done some research on animal experiments for uncovering its mechanism. These studies have proved the theory is very useful in clinic and supported the idea:the most important thing for facilitating spleen is invigorating it rather than replenishing it. But few people mentioned the Bile-Discharging Therapy on treatment of the children anorexia. Tutor found from much observation by the previous study that Spleen- Invigorating Therapy at the same time with Bile-Discharging Therapy on treatment of the children anorexia get better effect. To systematic observe the clinical efficacy of taking the Prescription of Spleen-Invigorating and Bile-Discharging on treatment of The Children Anorexia due to Disharmony of Spleen and Stomach; To chatacterize the molecular structural features of intestinal microbial communities of anorexia children before and after treatment. MethodsAdopt randomized controlled trial.60 cases of children which meet the diagnostic criteria according to the 1:1 proportion were randomly divided into two groups:the experimenral group and control group. The experimenral group took the Prescription of Spleen-Invigorating and Bile-Discharging (Poria 10g, Citrus 3g, Turmeric 6g, Capillaris10g,Magnolia flower 6g, Lentil Flower 6g, Perrin 6g, Bupleurum 3g).The control group used Xiao er huashi oral liquid. Children of the two groups stoped using all drugs which affect the results of experiment during treatment except the drug of experimental program. Totol course is seven days. We recorded the Chinese Medicine syndrome scores, determined urine amylase. Before and after treatment, the total DNA of microbial communities in experimenral group children's fecal samples were amplified by using the ERIC primers to generate community finger-prints.Result1. Before and after treatment, the Chinese medicine syndrome scores of the experimenral group were 8.93±2.30 and 1.97±1.56 respectively; urine amylase was 156.20±127.07 and 231.40±133.45 respectively. Before and after treatment, the Chinese medicine syndrome scores of the control group were 8.57±2.16,3.47±1.07; urine amylase was 166.78±149.92,211.50±144.77. By t test, the experimenral group and control group could improve the Chinese medicine syndrome scores and urine amylase. Before and after treatment, the effects were significantly different. It also showed that experimenral group is more effect than control group in improving the Chinese medicine syndrome scores. But urine amylase between the two groups, no statistical signigicance. Experimenral group showed overall clinical control rate of 80% and the total effective rate was 96.7%;was better than 30% and 93.3% of control group. The result was statistically significant.2. Ten samples before and after treatment of experimenral group use the ERIC primers to generate stable and repeatable community finger-prints. Samples of No.13 before and after treatment of experimenral group could not amplified bands. So the samples were removed. So the samples of finger-prints before and after treatment of experimenral groups were 9 cases respectively. When the subjects were divided into two groups:before treatment of experimenral group (B) and after treatment of experimenral group (A), the total number of operational taxonomic unit (OUTs) represented by unique ERIC-PCR bands for each group was 25 and 29 respectively. The corresponding Shannon's diversity indexes were 1.456±0.271 and 1.650±0.462 respectively. Accumulative curves of Cs values of the two types of samples were plotted and the proportions of Cs lessthan 0.4 were 89% for B and 97% for A.Conclusion1. The Prescription of traditional Chinese medicine of Spleen-Invigorating and Bile-Discharging on Treatment of the children anorexia could improve the total Chinese medicine syndrome scores and urine amylase. It was more effect than Xiao er huashi oral liquid in improving the Chinese medicine syndrome scores. 2. The diversity of intestinal microbial communities of group of anorexia children before treatment has been significantly reduced compared with the group of anorexia children after treatment of the Prescription of Spleen-Invigorating and Bile-Discharging.
