1. BackgroundCerebrovascular disease is one of the most urgent, fatal and acute illnesses.Its dealth rate is 1000/10 million people world wide among them, cerebrovascular disease is 85%. Therefore, the pace of the ischemic-stroke will on the rise gradually due to the increasing number of high blood preasure patients and aging population. It is not onlu a serious threat to the patient's life but also it affects their quality of life. It's a heavy burden to the patient, family and society as a whole. So treatment of stroke have been a hot topic among the health profession in China and overseas. It has two mechanism-technical ereas. One is modern rehabilitated technique which include Physical Therapy, Occupational Therapy, Speech Therapy etc. The often is conventioanl acupuncture, such as body-acupuncture, scalp-acupuncture, and eye-acupuncture etc. Treating stroke, patients with manipulatia, techniques and abundant experiments has been well-established, accepted and widely-used in TCM field. To treat after-stroke patients with acupuncture is useful and effective especially to ischemic-stroke patients. It heeps to protect, repair and manipulate its system. It has become a vital topic in the medical field. JAK/STAT signal transduction system has recently been discovered as a pathway in which it can activate cell increase, cell division, death and the process of immune manipulation. JAK/STAT protein not only reflects in the nevours system, but also plays a decisive role in the process of apoptisis in the focal region. This research paper is to study the focal cerebral-ischemia area of the rats model, during its acute, chronic period, its STAT3 changes and its level, and attempts to find out the function and expression of the nervous cells damage of the focal area.2. Methods MCAO is the way to make the model. Observing the changes level of STAT3 in the brain of cerebral isehemia rat and changes of behavior in 2hours,1 day,3 days,1 week,3 weeks after injury, as well as electro-aeupuneture Baihui(DU20), Dazhui(DU14), find out the impact of these indicators and to explore the needle mechanisms.3. The results3.1 The changes of neurofunetional scores after elctro-apuncturing in focal cerebral ischemia rats.Sham-operated group throughout all periods, the malfunctions or the nervous cell damages was zero, there was no difference among groups(P>0.05). Ischemia-stroke in 1-day period, the rats'brain nerve cell damges average was higher between the model group and elctro-acupuncture(EA) group. The score of the model group got the highest. The rats slided from the ischemic were to the healthy side or maked circle. There was a significant difference when compared with the EA group(P<0.01). It shows that using EA treatment was remarkable and the ischemic behavious have been improved tremendously. Ischemic-stroke in 3-day period, the nerve cell damages have been reduced between the model group and EA group, the restoration rate in the EA group was much faster than the model group. The difference was obvious(P<0.001). This indicates that EA tackled the focal ischemic-stroke area and restores its function with a positive impact. Obviously, there was quite an outstanding difference between 1-day and 3-day periods of the EA groups(P<0.001). Ischemic-stroke in 1-week period, there was improvement by average in all groups, the average score was lower than the 3-day group. The gap between the model group (1-day) and EA Group (1-week) was easy to see(P<0.05). The difference was substantial between the EA group(1-day), EA group(3-day),and EA group (1-week). This meant that the more time using the EA, the between in terms of its effectiveness in improving the nerve cell function in the problem area. Ischemic-stroke in 3-week period, nerve cell dysfunction delines in both the model group and EA group, the difference between the model 3-week groups and 1-week group was quite significant, this demonstrates that there was self-repair mechamism in the brain. But there was no difference between the EA 1-week group and the 3-week group. This is to prove that chronic ischemic-stroke doesn't respect to the EA treatment.3.2 The changes of STAT3 after apuncturing in focal cerebral ischemia rats. There was a close relationship between STAT3 changes and ischemic stroke, therefore, to study STAT3's signal transduction system, manipulation and EA's intervention after stroke offers meaningful insight in how they work in the brain. The result illustrates that the sham-operated groups'STAT3(OD) in the 2-hour group,1-day group,3-day group,1-week group and 3-week group, there was no significant difference. Statistically, there was no apparent difference(P>0.05).Ischemic-stroke in 2-hour period for model and EA group, there's an increase in STAT3, the STAT's OD is higher than the sham-operated group(P<0.05 and P<0.001 respectively). Snd there was no ovbious difference between the two groups(P>0.05); this demonstrares that ischemic-stroke 2-hour period, STAT3 in the focal area increases, this means EA's effect on the STAT3 is invalid.Ischemic-stroke in 1-day period, the STAT3 level increasd tremendously for both model group and EA group. The OD was quite different with the sham-operated group by comparison(P<0.001 for both groups). There was certain difference between them(P<0.01). The STAT3 level was signifficant between the model group (1-day) and 2-hour group (P<0.001) when compared with EA (1-day) and 2-hour group, the difference was obvious(P<0.05). This indicates that ischemic-stroke in 1-day period, STAT3 in the focal area will increase. Significantly, it meant apoptosis has reached its peach. STAT3 might have signalled in the process of the apoptosis. EA can definitely lower its level. This explaims that EA enhance its protection mechanism and plays an important role in the brain.Ischemic-stroke in 3-day period, the STAT3 levels of model group and EA group were seen in different degrees of decline, there are still significant differences in the level of two groups in OD compared with the sham-group (both P<0.001). Data show that EA(3-days)'s STAT3 level is lower than the model, but both shows no significant difference between (P> 0.05); model group (1-day) and model(3-days) group significantly different (P<0.01); EA(3-day) group and EA (1-day) group are significantly different between groups (P<0.05). Tip: 3 days after cerebral ischemia, the level of STAT3 in electro-acupuncture group and model group decreased, which also showed a decline in apoptosis. The data showed the STAT3 level of EA group decreased lower than the model group, but no significant difference between the two. That cerebral ischemia 3 days later, the rat brain by reducing the level of STAT3 protein in order to participate in their own protective mechanisms, electro-acupuncture treatment can promote this process to some extent, the role of protection mechanisms in the brain.Ischemic-stroke in 1-week period, the model group and EA group were seen STAT3 levels continue to decline, model (3-day) group and model (1-week) group, EA (1-week) group and EA (3-day) group between the two groups were significantly different (P both P<0.001). Compared mode (1-week) group to model (1-day) group, model (3-day) group, there were significant differences (both P<0.001); In EA group, the EA (1-week) group and the EA (2-hour) group, EA(1-day) group and EA(3-days) group, all existed significant group differences (all three P<0.001); However, from the data point of view, the STAT3 level of EA (1-week) group was slightly lower than in model (1-week) group, but the difference of values between the two was no significant difference (all P>0.05). Tips, Ischemic-stroke in 1-week period, the STAT3 levels of model group and EA group continue to decline, while no significant difference between the two groups, suggesting that the brain self- protective mechanism exists, the downward effect of electroacupuncture on STAT3 have some continuity.Ischemic-stroke in 3-week period,, there was no significant change in the level of STAT3 in model group, but STAT3 levels of EA group decreased continuously. There was no significant difference in the OD values of two groups of STAT3 compared with the sham-operation (both P>0.05); model (3-weeks) group and model (1-weeks) group also showed no significant difference between groups (P>0.05); Compared EA(3-week) group and EA(1-week) group there was significant difference (P<0.05); Model (3-weeks) group and EA (3-weeks) group existed significantly different (P<0.05). Tips:cerebral ischemia 3 weeks is chronic stroke, STAT3 levels of EA group continued to decline, which had significantly different with the model group.It indicates that the accumulation of electrical stimulation to improve the effect on brain function may be related to the decline of STAT3. There is a certain positive effect. Using electro-acupuncture in the treatment of ischemic stroke in the chronic phase.4. ConclusionIn a word, localized ischemic-stroke can lead to paralysis and a series of nerve cell damages syndromes. EA treatment can possibly improve the nerve cell's function of the brain in rats, it tackles the internal mechanism and its relationship with manipulation and the STAT3 protein, that means to lower the STAT3 level expression with this method. EA is able to lower the STAT3 protein level and heops repair the ischemic-stroke brain damages, this will have a great impact treating the stroke.
