Research on the chemical studies of two compounds extracted from the root of Peucedunum harry-smithii var subglabrum,We established method for analysis of the compounds of Pharmacokinetic studies.The dried root of Peucedanum as an important Chinese herbal medicine,is used to dispel wind and remove heat, relieve cough and resolve phlegm.There are several plants of Peucedanum which have been used as the important traditional Chinese medicine "qianhu" besides Peucedunum praeruptorum Dunn.Peucedunum harry-smithii var subglabrum,known as shaomaobeiqianhu in the northwest region of China,is an important species of Peucedani.50,The contents of Pteryxin and Bergapten have many pharmacological activities.The potentially medically valuable pyranocoumarin Pteryxin and Bergapten were isolated from the root of Peucedanum harry-smithii var subglabrum by chloroform and then partitioned by chromatography.The structures were rigorously proved by comparision with standard products.To develop a LC-MS-MS method to determine Pteryxin concentration and its metabolite in rats plasma.Blood samples were separated on a Hypersil ODS column (4.6 mm×250 mm,5μm),The mobile phase consisted of methanol:water (70:30, v/v),The flow rate was 1 mL/min.The lower limit of quantification was 0.1325μg·L-1,the lower limit of detection was 0.053μg·L-1,the extraction recoveries were more than 83% (n=5),respectively.The inner and inter-days precision were both less than 10%.The method was set up and adapted for the analysis of small biological samples taken from rats.We have established a UPLC-MS-MS method for analysis of Pteryxin in mice plasma and tissue,using isoimperatorin as internal standard,samples were separated on a BEHC18 column (1.7μm,2.1×50mm),The mobile phase consisted of methanol:water (70:30, v/v). The flow rate was 0.2mL/min.The lower limit of quantification was 1.54 ng·L-1,the lower limit of detection was 7.7 ng·L-1,The results presented here clearly indicate that this proposed method could be applicable to investigate the pharmacokinetic and tissue distribution of Pteryxin in mice after administration.To develop a LC-MS-MS method to determine Bergapten concentration in rats plasma. using isoimperatorin as internal standard,samples were separated on a Hypersil ODS column (4.6×250 mm,5μm),The mobile phase consisted of methanol:water (77.5:22.5, v/v). The flow rate was 0.8 mL/min. The lower limit of quantification was 8.12μg·L-1.The method described in this report has high sensitivity and selectivity.
