Objective:Radix Peucedani is the root of the umbelliferous plant of Peucedanum praeruptorum, which is recorded in the Chinese pharmacopoeia for 2010 edition. Peucedanum praeruptorum resources are rich and widely distributed in China,which are produced mainly in the provinces of Zhejiang,Henan,Hunan,Sichuan and so on. Radix Peucedani is bitter and pungent to the taste,as well as slightly cold.It has the function of evacuating wind heat, reducing gas and resolving phlegm.Therefore,it is used as one of the widely traditional Chinese medicinal herb for bronchitis and trachea of wind heat,cough, excessive phlegm and asthma.Through the modern pharmacological research, Radix Peucedani is proved to have certain effects of bronchitis, tblood pressure, coronary heart disease and so forth. Furthermore,it has the broad application prospect for treating cardiovascular diseases. Peucedanum harrysmithii var. subglabrum is originated from the umbelliferous plant of Peucedaneae drude plant, which are produced mainly in the regions of Longnan and Tianshui of the province of Gansu,as well as the eastern area of this province.It is widely used as the local traditional Chinese medicinal herb. This paper is aimed to set up the the high performance liquid chromatography (HPLC) fingerprint of the various extract part of Peucedanum harrysmithii var. subglabrum.Then the experiments of antitussive action and reducing phlegm function are conducted.By the the grey relational analysis, the relationship between the HPLC fingerprint of the various extract part of Peucedanum harrysmithii var. subglabrum and its optimal drug effect is investigated.Meawhile,the correlation values of antitussive action for water extraction and reducing phlegm function for chloroform extraction are calculated repectively.The contribution values of drug effect are found by the correlation values.Ultimately.the characteristic peaks are established for drug effects.Method:The four extraction of 10 batches of Peucedanum harrysmithii var. subglabrum from various Gansu regions were analysed by HPLC. Then the the HPLC fingerprint of the various extract part of Peucedanum harrysmithii var. subglabrum and its common mode were set up. Using the chromatographic fingerprint of computer aided similarity evaluation system software,the HPLC fingerprint of Peucedanum harrysmithii var. subglabrum was evaluated by similarity.The antitussive action and reducing phlegm function of various extract part were compared respectively.Through obtaining the quantized basis of the HPLC fingerprint and drug effects as well as using the grey relational analysis, the drug effect contribution values of chemical compositions were found from the characteristic peaks of the HPLC fingerprint.Results:The 10 batches of HPLC fingerprint of Peucedanum harrysniithii var. subglabriim for the chloroform extraction and ethyl acetate extraction had 6 reference substances,which were marmesin, psoralen, xanthotoxin, Pd-Ib, pteryxin, and Peuformosin,and the content of pteryxin was highest.Nonetheless,its HPLC fingerprint for ethanol extraction and water extraction had not such reference substances above.The similarity of 10 batches of Peucedanum harrysniithii var. subglabrum for four extractions were more than 0.9. Subsequently,the experiments of antitussive action and reducing phlegm function for various extractions were conducted.We could find that the chloroform had the remarkable reducing phlegm function and the water extraction had the remarkable antitussive action.By the grey relational analysis, the contribution order of reducing phlegm was (number of peaks) 13>12>16>18>19>6>20>14>1>11> 15>10> 17>2>5>4>7>3>8>9, in which the number of peaks of 1,3,4,10,13 and 16 were identified as marmesin. psoralen, xanthotoxin, Pd-Ib, pteryxin, and Peuformosin; the contribution order of antitussive action was (number of peaks) 10>5>7>1>8>9>2> 4>3>6.Conclusion:The study establishes a new comprehensive quality control mode to integrate the spectrum-effect relationship.and provides a theoretical basis of further research for Peucedanum harrysmithii var. subglabrum.Thus it has a certain application value.
