Objective We examined the distribution of single nucleotide polymorphisms(SNPs) rs2746342 of amp-activated protein kinase(AMPK) gene among gansu population with type 2 diabetes (T2DM) and noumal population,in order to investigate the association between the polymorphisms of human AMPK-rs2746342 gene and type 2 diabetes,Adiponectin,Resistin.Methods 163 cases of type 2 diabetic patients and normal controls 86 patients,Fasting blood was collected,Genomic DNA was isolated from peripheral blood leucocyte according to protease K method. Applying means of polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP),the polymorphism of AMPK- rs2746342G/T were determined. By means of ELISA to examine the level of Adiponectin,Resistin.Results (1)There was polymorphism of AMPK- rs2746342 in both type 2 diabetic patients and normal controls patients.The distribution of AMPK- rs2746342T/T,G/T,G/G genotypes: type 2 diabetic patients group was 35%,53%,12%,respectively, T allele frequency was 61.5%; normal controls group was 31%,43%,26%, T allele frequency was 52.5%; (2) The level of adiponectin in type 2 diabetes with obesity is lowerer than normal weight in type 2 diabetes and normal controls (p (0.05), The level of resistin in type 2 diabetes with obesity is higher than normal weight in type 2 diabetes and normal controls(p<0.05); (3) The level of TG and LDL in type 2 diabetes with obesity is higher than normal weight in type 2 diabetes and normal controls(p (0.05), HDL is lower(p<0.05); (4)Correlation analysis makes clear: adiponectin and BMI,BP are negative correlation, adiponectin and F-CP is positive correlation (p<0.05), resistinand BMI,BP are positive correlation, resistin and F-CP is negative correlation (p<0.05); (5) Resistin,TG,CH,LDL,F-CP is in lower, Adiponectin,HDL isin higher in the genotype T/T, G/T, G/G of Type 2 diabetes groups(p<0.05).Conclusion (1) The polymorphism of AMPK- rs2746342G/T existed among gansu population,and it was revelant with T2DM,the G/G gene frequency of Type 2 diabetes group is lower than normal control guoup,according to this,G allele maybe the protecting gene, T allele maybe the risk gene; (2)There was significant association between the polymorphism of AMPK- rs2746342 and adiponectin and resistin.The adiponectin is higher, the resistin is lower in G/G gene type.According to the F-CP is higher in T/T gene type,means rising insulin resistance probanly, BP,TG,CHO,LDL is higher, HDL is lower, maybe revelant to type 2 diabetes complications.G/G gene type is opposite,maybe protect the patient of type 2 diabetes.
