The Effects Of Shuxuetong Injection On Group Ⅱa Secretory Phospholipasea2 Expression In Aorta Of Rats With Atherosclerosis

Background:Coronary heart disease is one of the major diseases which threat to human health, In recent years, the prevalence of coronary heart disease increased significantly.With living standards improving and lifestyle changing, patients gets younger and younger. Currently, the pathogenesis of coronary heart disease has become hot research field of cardiovascular disease. More and more scholars accepted the formation of atherosclerotic inflammation proposed by Ross. Many studies have shown that secretory phospholipase A2 has close relation with the formation and instability of atherosclerosis plaque, sPLA2Ⅱa is an independent risk factor for coronary heart disease.Shuxuetong injection, with the role of anti-clotting, anti-thro mbosis and solubling fibrinogen, is extracted and refined from Chinese herbal medicines such as leech and earthworm. Clinical practice shows that Shuxuetong had a better role in atherosclerosis. So it is widely used in coronary heart disease, cerebral infarction and other cardiovascular and cerebrovascular disease . There is no basic research of traditional chinese medicine reported on sPLA2 IIa at home and abroad.Objective:To establish a rat model of atherosclerosis, observe the expression of group IIa secretory phospholipase A2(sPLA2 IIa) in atherosclerosis model rats'aorta. Exploring possible impact and mechanism of Shuxuetong injection on sPLA2Ⅱa.Method: Thrity healthy Wistar rats were randomly divided into three groups:normol group( n = 10),model group ( n = 10),and shuxuetong group(4ml·kg-1·d -1) ( n = 10).The rats of normal group were fed with normal diet .Those of model group were fed with high fat diet. Those of Shuxuetong group fed with high fat diet and accepted intravenous injection of Shuxuetong Injection (4ml·kg-1·d -1) . At the end of 12 weeks, we observe the blood fat and blood sugar level of rats in each group.We detected the expression of sPLA2 IIa in rats aorta in each group by immunocytochemistry and western blot method.Results: sPLA2 IIa expression was mainly located in the aortic wall smooth muscle cells and amorphous material of atherosclerotic plaques. Compared with the normol group, the expression of sPLA2 IIa in the aortic wall in model group was significantly increased (P <0.01);Compared with model group, Shuxuetong sPLA2 IIa injection group was significantly reduced (P <0.01).Conclusion:The expression of sPLA2-IIa may be one of important reasons in the pathogenesis of atherosclerosis. Through reducing the expression level of sPLA2-IIa in aorta ,Shuxuetong injection could relieve the process of atherosclerosis .
