ACE2 Inhibited The Expression Of Angiotensin â…¡ Type 1 Receptor MRNA In Vascular Smooth Muscle Cells Of Spontaneously Hypertensive Rat.

AIM:To investigate the effects of ACE2 gene transfection on the expression of Angâ…¡type 1 receptor mRNA in VSMCs of SHR.METHODS:The pm-ACE2 was transfected into VSMCs of SHR by lipo-fectamine 2000. VSMCs were devided into normal cell group,pcDNA3.1/Hygro(+) transfected group,pm-ACE2 transfected group,irbesartan group,pm-ACE2 transfected + irbesartan group. Detecting the expression of AT1 receptor mRNA through using RT-PCR.RESULT: The OD value of AT1 receptor mRNA of ACE2 transfected group were obviously lower than that of normal cell group and pcDNA3.1/Hygro(+) transfected group, the effect was concentration-dependented(4ug-10ug)(P<0.05) and time-dependented(12h-72h)( P<0.05). The effect of irbesartan inhibited the expression of AT1 receptor mRNA in VSMCs ofSHR was concentration-dependented (10^-9 mmol/L-10^-5mmol/L) ( P<0.05) and time-dependented (2h-24h)( P<0.05) . To campare with pm-ACE2 transfected group and irbesartan group,the OD value of pm-ACE2 transfected + irbesartan group was obviously decreased(P<0.05).CONCLUTION:The expression of AT1 receptor mRNA can be inhibited through transfecting pm-ACE2 into VSMCs of SHR, the effect was time-dependented and concentration-dependented. Irbesartan inhibited the expression of AT1 receptor mRNA in VSMCs of SHR.ACE2 and irbesartan have synergistic effect on attenuating the expression of AT1 receptor mRNA in VSMCs of SHR.
