AIM:To investigate the effects of ACE2 gene transfection on expression of AngⅡtype 1 receptor mRNA in vascular smooth muscle cells of SD rat.METHODS:Transfect pm-ACE2 into vascular smooth muscle cells of SD rat by lipofectamine 2000. VSMC were devided into normal cell group,pcDNA3.1/Hygro(+) transfected group,pm-ACE2 transfected group,AngⅡgroup,pm-ACE2 transfected + AngⅡgroup. Detecting the expression of AngⅡtype 1 receptor mRNA through using RT-PCR.RESULT: The OD value of AngⅡtype 1 receptor mRNA of ACE2 transfected group were obviously lower than that of normal cell group and pcDNA3.1/Hygro(+) transfected group, the effect was time dependence (8h—72h)(P<0.05)and concentration dependence(6ug—8ug)(P<0.05). The effect of AngⅡon expression of AngⅡtype 1 receptor mRNA was concentration dependence .When the concentration of AngⅡwas lower than 10-8mol/L ,the expression of AngⅡtype 1 receptor mRNA was higher than that of normal cell group and pcDNA3.1/Hygro(+) transfected group. The OD value of pm-ACE2 transfected + AngⅡgroup was obviously lower than that of normal cell group and pcDNA3.1/Hygro(+) transfected group(P<0.05),but obviously higher than that of pm-ACE2 transfected group.CONCLUTION:The expression of AngⅡtype 1 receptor mRNA can be inhibited through transfecting pm-ACE2 into vascular smooth muscle cells of SD rat, the effect was time dependence (8h—72h)and concentration dependence(6ug—8ug). ACE2 can reverse the effect of AngⅡ.
