【Objective】To investigate the function of TNF-α, NF-κβin the role of liver ischemia-reperfusion injury, using immunohistochemistry with the model of hemorrhagic shock and reperfusion liver injury.【Methods】Thirty rats were randomly divided into sham operation group ( Group A ), shock group ( Group B), hemorrhagic shock / resuscitation (Group C). Anaesthetize the rats in Group A, when maintaining mean arterial pressure (MAP) is normal, directly collect liver tissue after the execution of rats. Puncture the femoral artery of Group B for bleeding,after MAP as (35±5)mmHg for 90 minitues,collect liver tissue of executed rats. Afterwards, anesthetize the rats in Group C, puncture the femoral artery for bleeding, then MAP as(35±5)mmHg for 90 minitues, recover the rats by reinfusion of autologous blood , maintaining MAP 80-100 mmHg to produce hemorrhagic shock and resuscitation model, moreover, collect liver tissue of executed rat after 6 hours of recovery. According to HE staining,the understanding of pathological changes of liver, the group condition of SP immunohistochemistry in liver TNF-α, NF-κβ,the regional integration absorbance of each group by automatic image analysis system (Image-proplus 6.0) can be gained. With the application of SPSS statistical software for data processing, apply the measurement data with the mean±standard deviation ( (x|-)±S), and use to compare among groups with t test,the statistic significance is obtained when P is less 0.05.【Results】HE staining of liver cell in Group A is normal in structure and morphology. Part of the vacuolar degeneration of liver cells with minor damage can be seen. Nevertheless, the heaviest damage in liver tissue is discovered in Group C, the hepatic lobule structure is unclear, the liver cells showed marked vacuole degeneration, in addtion, there is sinus congestion in liver whose plent of exudate and liver neutrophil accumulation of macrophages is visible. The results of immunohistochemistry showed: The highest TNF-αand NF-κβwhose average absorbance values were (0.197±0.026,0.131±0.035) exsits in Group C, while in Group B ,with (0.133±0.023,0.098±0.028),the average absorbance values are lower than Group C.Group A indicats the lowest positive expression with (0.075±0.030,0.045±0.021)in the average absorbance values. All of the above results are analyzed by SPSS software, using to check over each group,moreover, P values were <0.05 which reveals statistically significant differences.【Conclusions】By successfully established hepatic ischemia reperfusion injury model that HE staining of liver pathology hemorrhagic shock occurred in the liver ischemia-reperfusion injury,it can be inferred that after hemorrhagic shock, the high expression of TNF-αand NF-κβcould possibly induce ischemia-reperfusion injury.
