Objective: Endometriosis (EMs, referred to as endometriosis) is a common and popular gynecologic disease in women of the reproductive of endometriosis has been distinctly increasing and many experts have attended it. EMs is defined as the growth of endometrial tissue (gland and stoma) outside the uterine cavity that often results in a vast array of gynecological problems including pelvic pain, menstruation and infertility. But its pathogenesis is still unclear. Endometriosis (EM) is a kind of benign disease but having the tumor biology characteristic. Hypoxia-inducible factor-1 (HIF-1) plays an important role of regulation in the cells response to adaptation of the hypoxic micro-environment, opens the tumor blood vessels generation links. As the key factor of angiogenesis, VEGF speed up the formation of the new blood vessels. Oxygen was known as the most important factor in numerous regulation VEGF expression. The aim of this experiment through animal experiment is to investigate the expression of Hypoxia-inducible factor-1 and VEGF in ectopic and eutopic endometrium, in order to reveal the part pathogenesis of endometriosis, and the method of using Soothing liver regulating qi warming yang on rats treated, we can discuss the effect and function mechanism of the method.Method: 70 sanitary and healthy Wistar females rat with normal estrous cycle, 200±20g in body weight, 20 are divided into normal group and sham operated group (each group is 10), others are used Wang Yan-ping's method to duplicate the model of endometriosis. After 4 weeks,chose the 40 rats that duplicate model triumphantly and divided them into 4 group: low-dose group, high-dose group, Danazol group, model group. There were 10 rats in each group. Those rats of low dose and high dose groups were administrated in dose of 15.41g/kg, 61.76g/kg separately, once per day for 4 weeks. The rats of Danazol group were in dose of 100mg once daily. Model group, sham-operated group and normal group were given the same amount of distilled water. The end time after the treatment period, the choice of estrogen rats fasting eight hours, were decapitated, and aseptic out each ectopic endometriosis caesarean organization, the normal group, sham operated group take endometrial organization .Carried them out immunohistochemical staining to determine the content of HIF-1,VEGF in endometrial tissue; At the same time, Checked another part of the organization to determine the content of in HIF-1mRNA endometriosis.Result:1.HIF-1expression of in positive transplant ectopic endometrium glandular epithelium cells in the cytoplasm, individual ectopic endometrium interstitial with small expression. The expression of HIF-1 in model group was higher than normal group, between the two groups was significant differences (P<0.05); the expression of HIF-1 in model group was higher than sham-operated group, between the two groups was significant differences (P<0.05); the expression of HIF-1 in high-dose group and low-dose group were lower than model group, the two groups had significant differences with model group(P<0.05); the expression of HIF-1 in high-dose group was lower than western medicine group, between the two groups was significant differences (P<0.05); between low-dose and western medicine group, the expression of HIF-1 had no difference(P>0.05); high-dose group, low-dose and normal group had no difference(P>0.05); the expression of HIF-1 was no significant difference in high-dose group and low-dose group (P>0.05).2.The expression of VEGF in eutopic and ectopic endometrium is mainly in the glandular epithelium cells of cell plasma, mass among a small number .VEGF mRNA exits in glandular epithelium cells in the cytoplasm. The expression of VEGF in model group was higher than normal group, between the two groups was significant differences (P<0.05); the expression of VEGF in model group was higher than sham-operated group, between the two groups was significant differences (P<0.05); the expression of VEGF in high-dose group and low-dose group were lower than model group, the two groups had significant differences with model group(P<0.05); the expression of VEGF in high-dose group lower than western medicine group, between the two groups was significant differences (P<0.05); between low-dose and western medicine group, the expression of VEGF had no difference(P>0.05); the expression of VEGF in high-dose group, low-dose and normal group had no difference (P>0.05); the expression of VEGF had no significant difference in high-dose group and low-dose (P>0.05).Conclusion:1. Hypoxia-inducible-1 and VEGF play an important role in the expression of endometriosis occurrence.2. HIF-1 and VEGF in the uterus tissue over-active certain synergy; Anoxic environment is induced HIF-1, VEGF expression of collaborative raised one of the reasons.3. HIF-1 and VEGF may have played a synergetic action in the angiogenesis of endometriosis.4. Shuganliqiwenyanghuayu Chinese medicine can reduce the expression level of HIF-1,VEGF,it plays a role in treatment of endometriosis by adjusting the distant metastases HIF-1,VEGF lever.
