Objective: By exploring the risk factors,prevalence,and pathogenesis of Chinese women cervical cancer in Tangshan area,and by studying the relations of interleukin-10 gene polymorphism,human papillomavirus infection and cervical cancer,providing the prevention for cervical cancer and the basis for application of HPV vaccine in Tangshan.Methods: Through the form of retrospective clinical epidemiological questionnaire for 218 cases of Han patients in tangshan,there were 103 cases of cervical cancer,115 cases of patients with non- cervical cancer were studied by clinical epidemiological investigation. The survey includes general information on past history,sexual and reproductive history,the history of contraception and smoking history,menstruation,marital status,by education degree,and so on.These factors in the control group and the case group have no significant difference,so the two groups are comparable. Extracting 3ml venous blood and take the cervical tissue about the size of 0.5 cm x 0.2 cm x 0.2 cm. Using DNA extracted kit extracted genomic DNA from serum and Phenolic chloroform extracted HPV-DNA from cervical tissue.Test the exists of IL-10-1082G/A and -592A/C in the above 218 patients'serum,and determine their genotype by ARMS-PCR and PCR-RELP respectively. Test the infection status of HPV16/18 in the above 218 patients'cervical tissue by PCR. Sending the PCR product of IL-10-1082G/A,-592A/C and HPV16/18 for sequencing.Using directly notation analysis haploid of IL-10 gene polymorphism site.Statistical analysising by SPSS13.0.Results: The result of the relations of interleukin-10 gene polymorphism,human papillomavirus infection and cervical cancer as follows:1 The relation of IL-10-1082G/A site gene polymorphism and cervical cancer Through analysis cervical cancer susceptibility and IL-10-1082G/A Site Genotype frequency and allele frequency: There are no significant differences in the control group and the case group(P>0.05).2 The relation of IL-10-592A/C site gene polymorphism and cervical cancerThere is no significant differences in the control group and the case group(P>0.05),but alleleA frequency is significant differences(P<0.05).3 The analyse of linkage disequilibrium between IL-10-1082G/A and IL-10-592A/C siteIt was found that haploid frequency of GC,GA,AA were 6.8%,18.0%,75.2% respectively in the case group,and 8.7%,27.0%,64.3% in the control group,and have a significant differences(P<0.05).4 The relation of HPV16/18 infection and cervical cancerThe differences of the infection rates of HPV16/18 and 16+18 in the case group and the control group were significantly(P<0.05).The infection rates of HPV16/18 and 16+18 in different pathological types were: Squamous cell carcinoma 69.2%,1.2% and 11.9%; adenocarcinoma 6.7%,0 and 46.7%;glandular squamous cell carcinoma only have one HPV16+18 mixed infection. HPV16/18 total infection rates have no significant differences in different ages(P>0.05). There is no significant differences in the relation between smoking and the infection of HPV16,HPV18,HPV16+18(P>0.05).5 IL-10-592A/C site gene polymorphism was different in HPV16/18 positive and negative groups,CA and AA gene type increased the susceptibility of HPV18(PCA=0.043,PAA=0.035).6 IL-10-1082G/A site alleleA was little different in HPV16/18 positive and negative groups,and increased the susceptibility of HPV16(P=0.049).Conclusion:1 interleukin-10 gene polymorphism have a relation with cervical cancer susceptibility,and IL-10-592A/C site alleleA may be the genetic susceptibility genes of cervical cancer,there is important significance to the occurrence, development and treatment of cervical cancer.2 In Tangshan area,it has a closely relation to cervical cancer with HPV16 and HPV16+18 infection,but HPV16 mainly causes Squamous cell carcinoma,HPV16 and HPV16+18 mixed infection mainly causes squamous cell carcinoma;ages and smoking have no relation with HPV16/18 and HPV16+18 mixed infection.3 In Chinese women in Tangshan area,there is a correlation between IL-10-592A/C site gene polymorphism and HPV16/18 infection,and IL-10 -1082G/A site alleleA is relation with cervical cancer caused by HPV16 infection.
