| Objective To study the expression of IL-23 mRNA and IL-17 mRNA on Endotoxin Induced Uveitis(EIU)at different time point and the relationship between the expression of IL-23 and IL-17 mRNA and ocular inflammationmanifestations .To investigate the possible pathogenesy of uveitis and to provide methods for preventing and treating uveitis.Methods 35 SD rats,female, 5~6weeks old,weight 180~210gwhich were eliminate eye diseases under the slim-lamp and opthalmoloscope。All rats were randomly divided into 7 groups(group A~G ) of 5 rats.Group B~G joint EIU model was induced by subcutaneous injection at two locations of E. coli LPS (100μg)+CFA(50ul).Group A joint control with subcutaneous injection at two locations of100ul PBS (pH 7.4).To observe the clinical manifestations on each model rat`s eyes and record scores every 2 hours. Models were killed randomly at 6h,12h,18h,24h,48h,60h after injection of E. coli LPS and control rats were killed at 24h after injection.Taken uveal retina from both eyes and frozen in liquid nitrogen.All samples were measured on the day of sample collection.To detect the espression of IL-23mRNA and IL-17mRNA by RT-PCR and did correlation analysis between IL-23/17mRNA and inflammation score .Results 4h after endotoxin injection ,the EIUgroup can be seen the iris blood vessels to dilate under the microscope , 6h shows mild anterior chamber flare, increased vascular congestion, edema, iris, eye inflammation score (1.4±0.16). 18 ~ 24h shows miosis; iris a high degree of edema, a serious charge vasodilatation, exudation, Thydall` s (+), partially visible hyphema, ocular inflammation score the highest (5.4±0.44). 24h after the symptoms gradually improved, the incidence rate was 100%. Control rats at different time points no significant change in ocular inflammation. The expression of IL-17mRNA,IL-23p19 mRNA and IL-12/23p40mRNA in the uveal tissue of experimental rats higher than control group (P <0.05), the difference was statistically significant. Correlation analysis of ocular inflammation scores and IL-17mRNA expression was positively (r17 = 0.88, P <0.05). Inflammation score and IL-23 p19/p40mRNA expression also positively correlated (rp19 = 0.96, rp40 = 0.83, P <0.05).Conclusion After endotoxin induced uveitis in SD rats eye occurs in the natural course of disease, RT-PCR detected the expression of IL-17 mRNA and IL-23 mRNA is increased .In the pathogenesis of uveitis may be associated with cytokine IL-17 and IL-23 related. The change and the severity of ocular inflammation were positively correlated suggesting that the severity of clinical symptoms of uveitis may be affected by of cytokines IL-17 and IL-23. |
